摘 要:RNA的提取是现代分子生物学中的一项常用技术,也是反转录、基因克隆、转录组分析等众多技术得以实现的基础。葡萄植株,特别是其成熟的果实、器官和组织中富含大量的糖、有机酸和酚类物质,这给高质量葡萄RNA的提取带来了极大的困难。本研究对近年来国内外研发的葡萄总RNA提取的方法进行了筛选和归纳,介绍了三种常见的葡萄RNA的提取方法:SDS/酚法、CTAB提取法和试剂盒法,对其利弊进行了对比和总结。分析表明,尽管适用于葡萄RNA提取的方法有很多,但其在葡萄总RNA提取中的效果却各有不同,试剂盒a提取出的RNA质量更高,改良SDS法可以提取出纯度高且量大的低分子量RNA,而SDS和CTAB法相结合的提取方法适合葡萄各种组织的RNA提取。RNA extraction is a common technology used in modern molecular biology,which is also the basis of many technologies such as reverse transcription,gene cloning,transcriptome analysis and so on.Grapevines,especially their mature fruits and other organs and tissues,are rich in sugar,organic acids and phenols,which makes it difficult to extract high quality total RNA from grapes.This study screened and summarized the methods for extracting total RNA from grapes developed at home and abroad in recent years,and introduced three common methods for extracting grape RNA:SDS/phenol method,CTAB extraction method,and kit method,and compared and summarized their advantages and disadvantages.The results showed that although there are many methods suitable for grape RNA extraction,their effects in grape total RNA extraction are different.The RNA extracted by kit a is of higher quality,and the improved SDS method can extract high purity and quantity.Large low-molecular-weight RNA,and the modified SDS method can extract low-molecular-weight RNA with high purity and large amount,and the combined extraction method of SDS and CTAB method was suitable for the extraction of RNA from various tissues of grapes.
