摘 要:目的观察转化生长因子β1(TGF-β1)基因缺失后中波紫外(UVB)辐射对皮肤组织的影响。方法制备TGF-β1基因缺失小鼠,使用UVB辐射制备皮肤损伤模型。实验分组为野生型设为对照组(Con组),野生型给予UVB照射设为Con+UVB组,TGF-β1-/-给予UVB照射设为TGF-β1-组。根据是否给予野生型小鼠TGF-β1注射,分为TGF-β1注射组TGF-β1(+)和TGF-β1不注射组TGF-β1(-)。分别观察小鼠皮肤损伤部位Smad3、基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-3(MMP-3)、Ⅰ型胶原及Ⅲ型胶原的mRNA表达情况,观察小鼠血清中类胰岛素生长因子-1(IGF-1)、血管内皮生长因子(VEGF)以及角质细胞生长因子(KGF)的含量。同时给UVB辐射皮肤损伤的野生型小鼠注射TGF-β1,观察MMP-1、MMP-3、Ⅰ型胶原及Ⅲ型胶原的mRNA表达情况,及血清中IGF-1、VEGF、KGF的含量。结果Con+UVB组Smad3表达和Ⅰ型胶原及Ⅲ型胶原含量低于Con组,血清中IGF-1、VEGF以及KGF含量低于Con组,而MMP-1和MMP-3的表达高于Con组,差异有统计学意义(P<0.05);TGF-β1-组Smad3表达和Ⅰ型胶原及Ⅲ型胶原含量低于Con+UVB组,血清中IGF-1、VEGF以及KGF含量低于Con+UVB组,而MMP-1和MMP-3的表达高于Con+UVB组,差异有统计学意义(P<0.05);TGF-β1(+)组Ⅰ型胶原及Ⅲ型胶原含量高于TGF-β1(-)组,血清中IGF-1、VEGF以及KGF含量高于TGF-β1(-)组,而MMP-1和MMP-3的表达低于TGF-β1(-)组,差异有统计学意义(P<0.05)。结论TGF-β1基因缺失会加剧UVB辐射对皮肤的损伤,给予TGF-β1补充可减少UVB辐射对皮肤的损伤。Objective To observe the impacts of ultraviolet B(UVB) radiation on skin tissue after TGF-β1 gene deletion. Methods The mice with TGF-β1 gene deletion were prepared, and the skin injury model was established using UVB radiation. Experimental groups were divided into the control group(the Con group, containing the wild-type), the Con+UVB group(containing the wild-type with UVB radiation), and the TGF-β1-group(containing the TGF-β1-/-with UVB radiation). The mice were divided into the TGF-β1(+) group(with TGF-β1 injection) and the TGF-β1(-) group(without TGF-β1 injection) according to whether or not wild-type mice were injected with TGF-β1. The mRNA expressions of Smad3, MMP-1, MMP-3, type Ⅰ collagen and type Ⅲ collagen at the skin injury site of mice were observed respectively, and the serum contents of insulin-like growth factor-1(IGF-1), vascular endothelial growth factor(VEGF) and keratinocyte growth factor(KGF) were observed. At the same time,TGF-β1 was injected into the wild-type mice with skin injury induced by UVB radiation to observe the mRNA expressions of MMP-1, MMP-3, type Ⅰ collagen and type Ⅲ collagen, and the serum contents of IGF-1, VEGF and KGF. Results The expressions of Smad3 and the collagen contents of type Ⅰ and type Ⅲ in the Con+UVB group were lower than those in the Con group. The serum contents of IGF-1,VEGF and KGF were lower than those in the Con group. The expressions of MMP-1 and MMP-3 were higher than those in the Con group, with statistically significant differences (P < 0.05). The expressions of Smad3 and the collagen contents of type Ⅰ and type Ⅲ in the TGF-β1-group were lower than those in the Con+UVB group. The serum contents of IGF-1, VEGF, and KGF were lower than those in the Con+UVB group. The expressions of MMP-1 and MMP-3 were higher than those in the Con+UVB group, with statistically significant differences(P < 0.05).
