摘  要:为促进马铃薯银屑病的分子生物学检测,从内蒙古牙克石地区马铃薯库房中采集了疑似马铃薯银屑病的样品,进行形态学鉴定,并对马铃薯银屑病分子生物学检测方法进行了筛选。结果表明,经形态学观察,块茎样品病斑特征符合“GB/T 28093-2001马铃薯银屑病菌检疫鉴定方法”中描述的病斑特征及病原菌特征。进一步利用常规PCR和巢式PCR对疑似样品进行了检测,两种方法均可扩增出特异性目的条带。目的序列经过测序,其序列与CBS640.85菌株(MH861907.1)序列的相似度达到100%。经形态学及分子生物学鉴定,可以证明在内蒙古牙克石地区马铃薯库房中采集的疑似马铃薯银屑病样品确为马铃薯银屑病,同时也证明本研究中采用的常规PCR和巢式PCR方法均可用于马铃薯银屑病的检测。In order to promote molecular biology detection of potato silver scurf disease,in this study,the samples suspected to be potato silvers curf disease from a storage room in Yakeshi Area of Inner Mongolia were morphologically identified.And the molecular biology detection methods of silver scurf were screened.The results showed that the characteristics of tuber lesions conformed to the characteristics of lesions and pathogens described in"GB/T 28093-2001 Method for quarantine and identification of potato silver scurf disease"by morphological observation.These suspected samples were detected by conventional PCR and nested PCR,and both of them could amplify specific target bands.The target sequence was sequenced,and its sequence similarity compared with CBS640.85 strain(MH861907.1)reached 100%.The samples collected from Yakeshi Area of Inner Mongolia were identified as potato silver scurf disease by morphological and molecular biology methods.It also proved that both conventional PCR and nested PCR methods can be used for the detection of potato silver scurf disease.
