摘  要:目的检测氯胺酮滥用及自然戒断后大鼠前额叶皮质、海马、纹状体中AMP、ADP、ATP的含量变化,探索氯胺酮滥用对不同脑区能量代谢的影响。方法将30只SD大鼠随机分为3组,连续10 d腹腔注射给药,对照组注射生理盐水,给药组注射30 mg/kg氯胺酮,戒断组连续10 d注射30 mg/kg氯胺酮后戒断1周。取脑组织分离前额叶皮质、海马、纹状体,用高效液相色谱法检测各样本中AMP、ADP、ATP的含量。结果大鼠脑组织中AMP含量为138.93~271.41 nm/mL,ADP含量为22.55~30.49 nm/mL,ATP含量为5.12~7.32 nm/mL。与对照组相比,给药组大鼠前额叶皮质AMP、ATP浓度显著升高(p<0.05),纹状体中ATP浓度升高(p<0.05);戒断组大鼠三脑区中AMP、ADP、ATP均与对照组大鼠无差异。结论氯胺酮滥用可干扰前额叶皮质和纹状体能量稳态。Objective To explore the effects of ketamine abuse and withdrawal on the contents of adenosine monophosphate(AMP),adenosine diphosphate(ADP)and adenosine triphosphate(ATP)in the prefrontal cortex,hippocampus and striatum of rats measured by a high performance liquid chromatography(HPLC)method,so as to explore the effects of ketamine on energy metabolism in brain region.Methods 30 SD rats were randomly divided into 3 groups:the control group,administration group and withdrawal group,which were injected with saline or 30 mg/kg ketamine for 10 consecutive days(withdrawal group were drug-free for 1 week after ketamine treatment).Then rats were killed and brain tissues were isolated and the content of AMP,ADP and ATP in brain regions detected by HPLC.Result The contents of AMP,ADP and ATP in rat brain tissues were 138.93~271.41 nm/mL,22.55~30.49 nm/mL and 5.12~7.32 nm/mL respectively.Compared with the control group,AMP and ATP in prefrontal cortex and ATP in striatum of ketamine group have significantly increased(p<0.05).While,there was no significant difference between the withdrawal group and the control group.Conclusion Ketamine abuse interferes with energy homeostasis in prefrontal cortex and striatum.
