摘 要:为了增强喷砂酸蚀(SLA)牙植体的生物活性,促进其骨整合能力,对SLA牙种植体表面进行氧等离子体亲水处理,将重组人骨形态发生蛋白-2(rhBMP-2)溶于壳聚糖(CS)溶液后静电喷涂(ES)在其表面,利用扫描电子显微镜、水接触角测量仪和X射线光电子能谱仪对SLA牙种植体表面进行理化特性表征,选用小鼠C2C12细胞系评价种植体表面的细胞黏附、增殖和分化能力。实验结果表明:静电喷涂构建的CS涂层保留了微米级孔洞结构,改善了表面亲水特性,实现了活性因子rhBMP-2的可控释放;体外细胞实验证实植体表面引入的rhBMP-2活性因子、微纳结构和涂层特性(亲水、功能基团)不仅明显地促进了细胞的黏附和增殖,而且具有协同促进成骨的效果。In order to enhance the biological activity of sand blast and acid etching(SLA)dental implants and promote their osseointegration ability,rhBMP-2 in chitosan(CS)solution was electrostatic sprayed(ES)on the surface of SLA dental implant with subsquent plasma hydrophilic treatment.The physical and chemical properties of SLA surface were characterized using scanning electron microscope,water contact angle measuring instrument and X-ray photoelectron spectrometer.The biological properties in terms of cell adhesion,proliferation,differentiation ability on the implant surface were evaluated using the C2C12 cell line.From the results,the CS coating constructed by electrostatic spraying retained the micron-scale pore structure with improved surface hydrophilicity,and the rhBMP-2 were released in a controlled manner.The biological testing in vitro confirms that the effective concentration of rhBMP-2,the micro/nano structure,as well as the properties of the coating not only significantly promoted the adhesion and proliferation of cells,but also showed the synergistical effect of promoting osteogenesis.
