摘 要:目的构建耐5-氟尿嘧啶(5-fluorouracil,5-Fu)的人结肠癌SW620/Fu和LoVo/Fu细胞株,以建立稳定的体外实验模型。方法选取亲本人结肠癌细胞SW620和LoVo进行培养传代,通过药物逐步递增法建立体外5-Fu诱导的耐药细胞株,并对耐药细胞株进行短串重复序列(STR)分析;采用MTT法分别检测5-Fu对亲本株和耐药株的半数抑制浓度(IC_(50)),并计算5-Fu耐药指数(RI)。结果耐药细胞株与STR分析结果高度匹配,表明成功构建人结肠癌细胞5-Fu耐药细胞株SW620/Fu和LoVo/Fu。5-Fu对SW620和SW620/Fu细胞的IC_(50)值分别为58.17±3.54和443.71±14.27μmol/L;对LoVo和LoVo/Fu细胞的IC_(50)值分别为152.53±13.95和1049.54±70.51μmol/L(均P<0.05),SW620/Fu和LoVo/Fu细胞的耐药指数分别为7.63和6.88。结论药物逐步递增法成功构建了5-Fu耐药细胞株,为结肠癌耐药机制研究建立了平台,有利于逆转肿瘤耐药的研究。Objective To construct 5-fluorouracil(5-Fu)-resistant human colon cancer cell lines SW620/Fu and LoVo/Fu,and to establish a stable vitro experimental model.Methods The parental human colon cancer cells SW620 and LoVo were selected for culture and passage.5-Fu-resistant human colon cancer cell lines was established by step-by-step increasing the concentration of 5-Fu to parental SW620 and LoVo cells in vitro.Short-string repeat(STR)analysis of drug-resistant cell lines was carried out.The median inhibitory concentration(IC_(50))of 5-Fu against parental and resistant colon cancer cells was detected using MTT assay,and the 5-Fu resistance index(RI)was calculated.Results The drug-resistant cell lines were highly matched with the STR analysis results,indicating that the 5-Fu-resistant human colon cancer cell lines SW620/Fu and LoVo/Fu were successfully constructed.The IC_(50)values of 5-Fu in SW620 and SW620/Fu cells were 58.17±3.54 and 443.71±14.27μmol/L,respectively;the IC_(50)values of 5-Fu in LoVo and LoVo/Fu cells were 152.53±13.95 and 1049.54±70.51μmol/L,respectively(all P<0.05).The RI of the SW620/Fu and LoVo/Fu were7.63 and 6.88,respectively.Conclusion The 5-Fu-resistant cell line was successfully constructed by the drug step-by-step method,which established a platform for the study of the drug resistance mechanism of colon cancer,and was beneficial to the research on reversing the tumor resistance.
