摘  要:目的 分析胃癌新辅助化疗患者组织中微小RNA-145(miR-145)、E-钙粘蛋白(E-cadherin)表达情况与其化疗效果的关系,对其评估胃癌新辅助化疗效果的意义进行探讨。方法 选择2019年1月至2020年10月接受新辅助化疗的胃癌患者80例,化疗前取患者肿瘤组织,以荧光实时定量PCR技术(qPCR)检测miR-145的表达水平,免疫组织化学法(IHC)检测E-cadherin在胃癌组织中的表达。评价患者的新辅助化疗效果,记录化疗相关不良反应。结果 80例患者共完成237个化疗周期,治疗有效率86.25%(69/80)。胃癌组织中miR-145、E-cadherin的表达水平低于癌旁组织(P<0.01)。肿瘤组织miR-145、E-cadherin表达与新辅助治疗效果及不良反应发生有关(P<0.05)。肿瘤组织miR-145、E-cadherin表达呈正相关(r=0.8264,P<0.001)。结论 在新辅助化疗前检测胃癌组织中miR-145及E-cadherin表达对预测新辅助化疗效果有较好的临床意义。Objective To investigate the expressions of microRNA-145(miR-145)and E-cadherin in the tissues of patients with different molecular types of gastric cancer,and to explore the clinical efficacy of neoadjuvant chemotherapy.Methods Eighty patients with gastric cancer who were treated by neoadjuvant chemotherapy win our hospital from January 2019 to October 2020 were enrolled in the study.The cancer tissues of patients were collected before chemotherapy,and the expression levels of miR-145 were detected by fluorescence real-time quantitative PCR(qPCR),and the expression levels of E-cadherin were detected by immunohistochemistry(IHC).Moreover the clinical effects of neoadjuvant chemotherapy were evaluated,and the adverse reactions related to chemotherapy were observed.Results The 80 patients completed 237 cycles of chemotherapy,with the total effective rate being 86.25%(69/80).The expression levels of miR-145 in cancer tissues were significantly lower than those in adjacent tissues(P<0.05).Moreover the expression levels of miR-145 and E-cadherin in gastric cancer tissues were closely related with the effects of neoadjuvant therapy and the occurrence of adverse reactions(P<0.05).There was a positive correlation between the expression of miR-145 and E-cadherin in tumor tissues(P<0.01).Conclusion The detection of the expressions of miR-145 and E-cadherin in gastric cancer tissue before neoadjuvant chemotherapy has important clinical significance for predicting the effects of neoadjuvant chemotherapy.
