摘 要:以石仙桃为原料,考察液料比、温度、超声时间、乙醇体积分数和粒径大小对石仙桃总黄酮提取率的影响.采用Design-Expert 8.05b中的Box-Behnken实验设计原理,以提取率为响应值,探究各因素对提取率的影响及其相互作用,建立各因素与提取率间的五元二次方程模型;同时以抗坏血酸为阳性对照,采用清除DPPH·和·OH对石仙桃总黄酮进行体外抗氧化研究.结果表明,石仙桃总黄酮的最佳提取工艺为:粒径0.180 mm、8、65%的乙醇溶液、液料比25 mL/g、温度60℃下超声40 min.在该工艺下,响应面预测值为1.83%,实测平均值1.80%,相对误差为1.64%,验证了该模型拟合的可靠性.石仙桃总黄酮对DPPH·和·OH体外抗氧化研究表明:石仙桃总黄酮具有抗氧化活性,在一定的范围内与DPPH·和·OH的清除率存在量效关系,其对DPPH·和·OH清除率的IC_(50)值为0.11和0.083 mg/mL;为石仙桃总黄酮提取和抗氧化研究提供一定理论参考价值.Taking Pholidota chinensis Lindl as raw material,the present study investigated the effects on the extraction rate of total flavonoids by liquid-to-material ratio,temperature,ultrasonic time,ethanol concentration and particle size.The Box-Behnken experimental design principle in Design-Expert 8.05b was used,The extraction rate was taken as the response value,The influence of various factors on the extraction rate and their interaction were explored,and a five-element quadratic equation model between each factor and the extraction rate was established.At the same time,through takingVC as a positive control,The antioxidant of the extraction of total flavonoids from Pholidota chinensis Lindl by the method of scavenging DPPH·and·OH free radicals was studied.The results showed that the optimal process for the extraction of total flavonoids from Pholidota chinensis Lindl by response surface optimization was 0.180 mm particle size,65%ethanol solution,liquid-to-material ratio 25 mL/g,and ultrasonic for 40 min at 60℃.Under this process,the response surface prediction value was 1.83%,the measured average value was 1.80%,and the relative error was 1.64%,which verified the reliability of the model fitting.The antioxidant research found that total flavonoid had a certain anti-oxidant activity and there was adose-effect relationship between total flavonoids and the DPPH·and·OH within a certain range.The IC_(50) of clearance rates were 0.11 mg/mL and 0.083 mg/mL.It provided a certain theoretical reference value for the extraction and antioxidant research of total flavonoids of Pholidota chinensis Lindl.
