circ_0001955靶向miR-1243调控肝癌BEL-7402细胞增殖、凋亡及EMT的机制研究

摘　　要：目的:探讨circ_0001955调控miR-1243对肝癌BEL-7402细胞增殖、凋亡及上皮间质转化(EMT)的机制研究。方法:肝癌BEL-7402细胞分为si-NC组、si-circ_0001955组、miR-NC组、miR-1243组、si-circ_0001955+anti-miR-NC组、si-circ_0001955+anti-miR-1243组。实时荧光定量PCR(RT-qPCR)检测circ_0001955和miR-1243的表达水平;MTT检测细胞活性;流式细胞术检测细胞凋亡;Western blot法检测蛋白表达;双荧光素酶报告实验检测circ_0001955和miR-1243的靶向关系。结果:与癌旁组织比较,肝癌组织中circ_0001955表达水平升高,miR-1243表达水平降低(P<0.05)。抑制circ_0001955表达后,肝癌细胞BEL-7402活性降低,凋亡率升高,E-cadherin蛋白表达水平升高,Vimentin蛋白表达水平降低(P<0.05)。circ_0001955靶向调控miR-1243;miR-1243过表达后,细胞活性降低,细胞凋亡率升高,Vimentin表达水平降低,E-cadherin表达水平升高(P<0.05)。干扰miR-1243表达逆转了抑制circ_0001955表达对肝癌BEL-7402细胞增殖、凋亡和EMT的作用。结论:抑制circ_0001955表达可能通过调控miR-1243抑制肝癌BEL-7402细胞增殖及EMT,促进细胞凋亡。Objective:To explore the mechanism of circ_0001955 regulating miR-1243 on the proliferation,apoptosis and epithelial-mesenchymal transition(EMT)of liver cancer BEL-7402 cells.Methods:Liver cancer BEL-7402 cells were divided into siNC group,si-circ_0001955 group,miR-NC group,miR-1243 group,si-circ_0001955+anti-miR-NC group,and si-circ_0001955+anti-miR-1243 group.Real-time fluorescent quantitative PCR(RT-qPCR)was used to detect the expression levels of circ_0001955and miR-1243;MTT was used to detect cell viability;flow cytometry was used to detect cell apoptosis;Western blot was used method to detect protein expression;dual luciferase reporter experiment to detect the targeting relationship between circ_0001955 and miR-1243.Results:Compared with adjacent tissues,the expression level of circ_0001955 in liver cancer tissues was increased,and the expression level of miR-1243 was decreased(P<0.05).After inhibiting the expression of circ_0001955,cell activity was decreased,BEL-7402 cell apoptosis rate was increased,E-cadherin protein expression was increased,Vimentin protein expression was decreased(P<0.05).circ_0001955 targets and regulates miR-1243;after miR-1243 was overexpressed,cell activity was decreased,cell apoptosis rate was increased,Vimentin expression was decreased,and E-cadherin expression was increased(P<0.05).Interference with the expression of miR-1243 reversed the effect of inhibiting the expression of circ_0001955 on the proliferation,apoptosis and EMT of liver cancer BEL-7402 cells.Conclusion:Inhibiting the expression of circ_0001955 may inhibit the proliferation and EMT of liver cancer BEL-7402 cells by regulating miR-1243 and promote cell apoptosis.
