摘  要:【目的】探讨罗伊乳杆菌及嗜酸乳杆菌对去卵巢骨质疏松小鼠肠道菌群的影响。【方法】按是否去卵巢和经益生菌喂养将80只C57BL/6J雌鼠随机分为8组,每组10只,分别为假手术空白组、假手术罗伊乳杆菌组、假手术嗜酸乳杆菌组、假手术罗伊乳杆菌混嗜酸乳杆菌组、去卵巢空白组、去卵巢罗伊乳杆菌组、去卵巢嗜酸乳杆菌组、去卵巢罗伊乳杆菌混嗜酸乳杆菌组。利用16rRNA高通量测序,对各组小鼠肠道菌群分类学组成、Alpha多样性及Beta多样性进行分析对比。【结果】经益生菌喂养的小鼠肠道菌群组成结构发生了变化。与未经益生菌喂养的小鼠相比,在去卵巢各益生菌组中厚壁菌门、产芽胞菌纲下降;在仅嗜酸乳杆菌组中变形菌门、丙型变形菌纲、肠杆菌目增加;在除了去卵巢嗜酸乳杆菌组中疣微菌门、阿克曼菌科增加。小鼠经益生菌喂养后,其肠道菌群的丰富度明显提升(P<0.05)。小鼠去卵巢后,其肠道菌群的丰富度与多样性均明显下降(P<0.05)。相同的建模与益生菌喂养方式对肠道菌群的影响是相似且有规律的。小鼠的肠道菌群会因去卵巢和喂养益生菌而发生变化。小鼠的喂养益生菌相比去卵巢的干预方式,对肠道菌群的影响更大。【结论】罗伊乳杆菌及嗜酸乳杆菌喂养能改变去卵巢骨质疏松小鼠肠道菌群的组成,这种改变存在着一定的规律,这可以为研究益生菌、肠道菌群与骨质疏松的关系提供实验依据。【Objective】To investigate the changes of gut microbiota in ovariectomized osteoporosis mice fed with Lactobacillus reuteri and Lactobacillus acidophilus.【Methods】C57BL/6J female mice(n=80)were randomly divided into eight different groups based on treatments:sham surgery(sham),sham with Lactobacillus reuteri feeding,sham with Lactobacillus acidophilus feeding,sham with both Lactobacillus reuteri and Lactobacillus acidophilus co-feeding,OVX surgery(OVX),OVX with Lactobacillus reuteri feeding,OVX with Lactobacillus acidophilus feeding,OVX and both Lactobacillus reuteri and Lactobacillus acidophilus co-feeding.There were 10 mice in each group.Data of 16rRNA high-throughput sequencing were used to analyze and compare the taxonomic composition,alpha diversity index and beta diversity index of gut microbiota in each group.【Results】Mice fed with probiotics had changes in the composition of their gut microbiota.Compared to the mice fed without probiotics,Firmicutes and Erysipelotrichia decreased in the OVX with probiotics fed groups.Proteobacteria,gamma proteobacteria and Enterobacterales increased in lactobacillus acidophilus fed groups.Verrucomicrobiota and Akkermansiaceae increased except for group of OVX with lactobacillus acidophilus fed.After the mice were fed with probiotics,the richness of gut microbiota was significantly increased(P<0.05).Ovariectomy made the richness and diversity of gut microbiota decreased significantly(P<0.05).The same surgery modeling and probiotic feeding patterns had similar and regular effects on gut microbiota.Mice fed with probiotics had a greater effect on their gut microbiota than the ovariectomized intervention.【Conclusion】Lactobacillus reuteri and Lactobacillus acidophilus can change the composition of gut microbiota in ovariectomized mice with osteoporosis.There are certain rules to this change.This can provide experimental basis for studying the relationship of osteoporosis with probiotics and gut microbiota.
