摘 要:目的研究主动饮酒对中脑腹侧被盖区(Ventral tegmental area,VTA)GABA(γ-aminobutyric acid)神经元形态学特征的影响。方法将雄性SD(Sprague Dawley)大鼠按体重随机分为对照组、饮酒组。对照组正常饮水,饮酒组以间歇性主动饮酒模式饮用20%酒精28 d,饮酒量达到3.2 g/(kg·24 h)为酒精成瘾大鼠。将建模成功的饮酒组大鼠在戒断0 h检测的数据称为0 h组,在戒断72 h检测的数据命名为72 h组。采用免疫荧光计数VTA GABA神经元,采用Elisa测定大鼠VTA谷氨酸脱羧酶(Glutamic acid decarboxylase,GAD)含量,电镜下观察神经元形态变化。结果与对照组(135.90±9.92)个/mm^(2)相比,戒断0 h组GABA神经元计数(77.88±9.33)个/mm^(2)和戒断72 h组GABA神经元计数(78.63±9.50)个/mm^(2)均明显降低,具有统计差异(F_(2,23)=110.7,P<0.01),戒断0 h组和戒断72 h组相比,无统计差异(t=0.16,P>0.05);与对照组(46.92±3.27)ng/mL相比,戒断0 h组的GAD含量(32.21±4.79)ng/mL和戒断72 h组GAD含量(33.04±2.79)ng/mL均明显降低,具有统计差异(F_(2),27=49.45,P<0.01),戒断0 h组和戒断72 h组相比,无统计差异(t=0.47,P>0.05);对照组线粒体结构正常,神经纤维髓鞘结构完整,戒断0 h组与戒断72 h组神经元内线粒体肿胀变性、嵴断裂,神经纤维髓鞘结构松懈,密度降低。结论主动饮酒使大鼠VTA GABA神经元的形态发生变化,需要进一步研究以阐明这种变化的机制,以制定有效的干预和治疗方法。Objective To investigate the effects of active drinking on the morphological characteristics of GABA neurons in Ventral Tegmental area(VTA).Methods Thirty male Sprague Dawley rats were randomly divided into control group and drinking group.The control group drank normal water,and the drinking group drank 20%alcohol intermittently for 28 days,and the alcohol consumption reached 3.2 g/(kg•24 h).Immunofluorescence was used to count the number of VTA GABA neurons.Another 35 male SD rats were randomly divided into control group and drinking group.The levels of Glutamic acid decarboxylase(GAD)in VTA were determined by Elisa.Finally,20 male SD rats were randomly divided into control group and drinking group.The morphological changes of neurons in each group were observed under electron microscope.Results Compared with the control group(135.90±9.92)mm^(2),the number of GABA neurons in the 0h withdrawal group(77.88±9.33)mm^(2) and the 72h withdrawal group(78.63±9.50)mm^(2) were significantly lower,there was statistical difference(F_(2,23)=110.7,P<0.01).There was no statistical difference between the 0h withdrawal group and the 72h withdrawal group(t=0.16,P>0.05).Compared with the control group(46.92±3.27)ng/mL,the content of GAD in the 0h withdrawal group(32.21±4.79)ng/mL and the 72h withdrawal group(33.04±2.79)ng/mL were significantly decreased,showing statistical differences(F_(2,27)=49.45,P<0.01).There was no statistical difference between the 0h withdrawal group and the 72h withdrawal group(t=0.47,P>0.05).In the control group,the mitochondrial structure was normal and the myelin structure of nerve fibers was intact.In the 0h and 72h withdrawal groups,the mitochondria in neurons were swollen and degenerated,the crists were broken,and the myelin structure of nerve fibers was loose and the density of nerve fibers was decreased.Conclusion Active drinking leads to morphological changes in rat VTA GABA neurons.Further studies are needed to elucidate the mechanism of these changes and to develop effective interventio
