上皮性卵巢癌患者血清中CENPU、ErbB2基因表达与患者临床病理特征及预后的关系

摘　　要：目的分析上皮性卵巢癌患者CENPU、ErbB2基因表达与患者临床病理特征及预后的关系。方法选择上皮性卵巢癌患者95例,查阅患者病案信息,收集患者年龄、性别及肿瘤位置、大小、病理分级、组织类型等资料。抽取患者肘静脉血3 ml,离心处理后下血清,使用全自动生化分析仪检测淋巴细胞计数和中性粒细胞计数。抽取患者肘静脉血3 ml,进行外周血白细胞DNA提取,采用荧光定量分析法检测CENPU、ErbB2基因表达。随访1年,统计患者生存时间。分析上皮性卵巢癌患者CENPU、ErbB2基因表达与患者临床病理特征的关系,探讨者CENPU、ErbB2基因表达与预后的关系。结果相关性分析发现,上皮性卵巢癌患者CENPU、ErbB2阳性表达与肿瘤位置、肿瘤大小、病理分级、肿瘤分期、淋巴细胞计数、中性粒细胞计数有关(P<0.05),与年龄无关(P>0.05)。CENPU阳性表达患者平均生存时间(4.58±1.08)个月,阴性表达患者(8.68±1.05)个月。ErbB2阳性表达患者平均生存时间(4.69±1.12)个月,阴性表达患者(8.71±1.16)个月。CENPU、ErbB2阳性表达患者生存时间少于CENPU、ErbB2阴性表达患者(P<0.05)。结论上皮性卵巢癌患者CENPU、ErbB2基因表达与患者临床病理特征及预后关系密切。Objective To investigate the correlation of CENPU and ErbB2 genes with clinicopathological characteristics and prognosis in epithelial ovarian cancer.Methods Ninety-five patients with epithelial ovarian cancer were selected.Data including age,gender,tumor location,tumor size,pathological grades and histological types were collected through questionnaires and medical records.After centrifugation of elbow venous blood,serum lymphocyte count and neutrophil count were measured using automatic biochemical analyzer.Extraction of DNA in peripheral blood leukocytes was performed,and the steps included routine anticoagulation,centrifugation,washing,shaking and ovulation,thereafter,CENPU and ErbB2 gene expressions were detected by real-time quantitative polymerase chain reaction assay.The survival time was recorded during the one-year follow-up period.Then the correlation of CENPU and ErbB2 genes with clinicopathological characteristics and prognosis in epithelial ovarian cancer was discussed.Results Correlation analysis revealed that positive expression of CENPU and ErbB2 in patients with epithelial ovarian cancer was associated with tumor location,tumor size,pathological grade,histological type,tumor stage,lymphocyte count and neutrophil count(P<0.05),and not with the age(P>0.05).The mean survival time was(4.58±1.08)months for patients with positive CENPU expression and(8.68±1.05)months for patients with negative CENPU expression.The mean survival time was(4.69±1.12)months for patients with positive ErbB2 expression and(8.71±1.16)months for patients with negative ErbB2 expression.The survival time of patients with positive expression of CENPU and ErbB2 was less than that of patients with negative expression(P<0.05).The expression of CENPU and ErbB2 genes in patients with epithelial ovarian cancer was closely related to the prognostic survival time of patients(P<0.05).Conclusion CENPU and ErbB2 genes are closely related to clinicopathological features and prognosis of epithelial ovarian cancer patients.
