摘  要:建立一种准确快速测定鸡蛋中VB_(1)含量的高效液相色谱—荧光法。样品经0.1 mol/L HCl在120℃条件下水解20 min提取后,用乙酸钠溶液调p H至4.0,然后经碱性铁氰化钾衍生,采用Xbridge C_(18)色谱柱分离,以乙酸钠-甲醇(80∶20,V/V)为流动相,荧光检测器检测。该方法具有较高的灵敏度,检出限为0.015 mg/kg,定量限为0.050 mg/kg;具有较宽的线性范围:1~500 ng/m L,线性关系良好(R^(2)>0.999);在1.0,2.0和10.0 mg/kg三个质量分数水平下,具有较好的加标回收率:95.5%~107.8%。该方法简化了样品前处理过程,节省了人工和试剂,提高了效率和准确度,满足鸡蛋中微量VB_(18)的检测要求,适用于大批量样品的测定;该方法对鸡蛋营养学研究及消费者进行鸡蛋消费选择具有重要的理论价值和现实意义。An accurate and rapid determination of VB_(1)in eggs by high performance liquid chromatography-fluorescence(HPLC-FLD)method was established.After the sample was hydrolyzed by 0.1 mol/L hydrochloric acid under 120℃for 20min,and p H was adjusted to 4.0.Then,it was derived by alkaline potassium ferricyanide.The separation of targeted derivative was performed on a Xbridge C_(18)column with the mobile phase of sodium acetate-methanol(80∶20,V/V)by HPLC-FLD.The method had high sensitivity,and the limit of detection(S_(LOQ))and the limit of quantitation(S_(LOD))of VB_(18)were 0.015 mg/kg and 0.050 mg/kg,respectively.The results showed that VB_(18)had a good linear relationship in the linear range of 1-500 ng/m L with the good correlation coefficient(R^(2)≥0.999),and a good recovery rate of 95.5%-107.8%at hree different concentrations of 1.0,2.0 and 10.0 mg/kg t.This method simplified the sample pretreatment process,saved manpower and reagents,improved the efficiency and accuracy,met the detection requirements of trace VB_(18)in eggs,and it was more suitable to analyze large quantities of samples;The method had important theoretical value and practical significance for the study of egg nutrition and the consumption choices for eggs.
