摘  要:目的通过响应面法优化地黄多糖的提取工艺并考察其抗氧化活性。方法使用水提醇沉法提取地黄多糖,在单因素试验的基础上,以提取时间、料液比和提取温度为自变量,以地黄多糖提取率为响应值,采用响应面法优化地黄多糖的提取工艺。对比Sevage法与三氯乙酸法脱蛋白的效果,并通过测定地黄多糖对DPPH自由基、羟基自由基的清除作用及Fe3+还原力评估其体外抗氧化活性。结果地黄多糖的最佳提取工艺条件为:提取时间100 min,料液比1∶60(g∶mL),温度70℃;在最佳条件下地黄多糖提取率可达7.09%,相对误差为0.29%。脱蛋白结果表明,三氯乙酸法较Sevage法更适于地黄多糖脱蛋白。多糖的抗氧化试验结果显示,在多糖质量浓度为1.0 mg/mL时,DPPH自由基、羟基自由基的最大清除率分别为72.44%、72.97%,还原能力评估吸光度值为0.5904。结论优化了地黄多糖的提取工艺,地黄多糖有较好的体外抗氧化活性,为地黄多糖的进一步开发利用提供了理论支持。Objective Response surface methodology(RSM)was used to optimize the extraction process of Rehmannia glutinosa Libosh.polysaccharide(RGP)and its antioxidant activity was evaluated.Methods RGP was extracted by water extraction and alcohol precipitation.A RSM experiment was used to optimize the extraction procedure of RGP based on a single-factor experiment,with extraction time,solid-liquid ratio and temperature as independent variables and the extraction yield as response value.The deproteinization effects of Sevage method and trichloroacetic acid(TCA)method were compared,and the in vitro antioxidant activity of RGP was assessed by the scavenging activity on 1,1-diphenyl-2-trimethylphenylhydrazine(DPPH)radical,hydroxyl radical and Fe3+reducing power.Results The optimal process conditions for RGP were as follows:extraction time of 100 min,solid-liquid ratio of 1∶60(g∶mL)and extraction temperature of 70℃.The extraction rate of RGP was 7.09%with relative error of 0.29%under the best extraction conditions.TCA method was more suitable for RGP deproteinization compared with Sevage method.The results of antioxidant experiments of RGP showed that when the concentration of RGP was 1.0 mg/mL,the maximum scavenging rate of DPPH radical,hydroxyl radical and the absorbance values corresponding to their reducing ability were 72.44%,72.97%,and0.5904,respectively.Conclusion The extraction process of RGP was optimized,and RGP had good antioxidant activity,which could provide a theoretical support for its further development and utilization.
