CDK9、iASPP在子宫颈癌组织中的表达及临床意义

摘　　要：目的探讨细胞周期依赖性蛋白激酶9(CDK9)、p53凋亡刺激蛋白家族抑制成员(iASPP)在子宫颈癌中的表达情况以及在子宫颈癌发生发展中的意义。方法收集正常子宫颈组织21例(对照组)、子宫颈低级别鳞状上皮内病变组织39例(低级别组)、子宫颈高级别鳞状上皮内病变组织40例(高级别组)及子宫颈癌组织45例(癌症组),采用免疫组织化学方法检测CDK9、iASPP在各组子宫颈组织中的表达情况,分析CDK9、iASPP表达与子宫颈癌临床病理特征的相关性,分析子宫颈癌组织中CDK9、iASPP的表达相关性。结果CDK9在对照组、低级别组、高级别组及癌症组中的阳性表达率分别为19.4%、43.6%、55.0%、68.9%,差异有统计学意义(χ^(2)=15.46,P=0.001);iASPP在对照组、低级别组、高级别组及癌症组中的阳性表达率分别为28.6%、51.3%、65.0%、73.3%,差异有统计学意义(χ^(2)=13.37,P=0.004);在子宫颈癌组织中,CDK9和iASPP表达与子宫颈癌的临床分期、浸润深度和淋巴结转移有关(P<0.05),与分化程度和脉管浸润无关(P>0.05);在CDK9阳性表达的子宫颈癌组织中,iASPP的阳性表达率更高,差异有统计学意义(χ^(2)=4.059,P=0.044);CDK9和iASPP在癌症组中的表达相关性为正相关(r=0.355,P=0.017)。结论CDK9、iASPP在子宫颈癌的发生发展和侵袭发展中具有协同作用,CDK9可能通过iASPP分子通路发生作用;CDK9和iASPP有望成为子宫颈癌治疗的潜在药物靶点及子宫颈癌患者临床评估预后的监测指标。Objective To investigate the expression of cell cycle dependent protein kinase 9(CDK9)and inhibitor of apoptosis-stimulating protein of p53(iASPP)in cervical carcinoma and their significance in the occurrence and development of cervical carcinoma.Methods 21 cases of normal cervical tissue(control group),39 cases of low grade cervical squamous intraepithelial lesions(low-grade group),40 cases of high-grade cervical squamous intraepithelial lesions(high-grade group)and 45 cases of cervical cancer(cancer group)were collected.The expression of CDK9 and iASPP in cervical tissues of each group was detected by immunohistochemistry.The correlation between the expression of CDK9 and iASPP and the clinicopathological features of cervical cancer was analyzed,and the correlation between the expression of CDK9 and iASPP in cervical carcinoma was analyzed.Results The positive expression rates of CDK9 in control group,low-grade group,high-grade group and cancer group were 19.4%,43.6%,55.0%and 68.9%,respectively,and the difference was statistically significant(χ^(2)=15.46,P=0.001).The positive expression rates of iASPP in control group,low-grade group,high-grade group and cancer group were 28.6%,51.3%,65.0%and 73.3%,respectively,and the difference was statistically significant(χ^(2)=13.37,P=0.004).In cervical cancer,the expression of CDK9 and iASPP was related to the clinical stage,depth of invasion and lymph node metastasis(P<0.05),but not to the degree of differentiation and vascular invasion(P>0.05).In the cervical cancer tissues with CDK9 positive expression,the positive expression rate of iASPP was higher,and the difference was statistically significant(χ^(2)=4.059,P=0.044).There was a positive correlation between CDK9 and iASPP in cancer group(r=0.355,P=0.017).Conclusion CDK9 and iASPP play a synergistic role in the occurrence,development and invasion of cervical cancer and CDK9 may play a role through the iASPP molecular pathway.CDK9 and iASPP are expected to be potential drug targets for the treatment of cervical carc
