摘 要:通过酒石酸亚铁比色法对沙棘茶叶提取液中茶多酚含量进行测定,采用单因素试验、正交试验确定反应的最优显色条件,在此条件下在540nm处测定其吸光度,用没食子酸标准品对照绘制标准曲线,并采用精密度试验、稳定性试验、加标回收率试验对最优显色条件加以验证,结果显示该条件稳定、合理可行。结果表明:最优显色条件组合为蒸馏水4mL、酒石酸亚铁5mL、磷酸盐缓冲液pH7.5。茶多酚浓度在10-50μg/mL浓度范围内具有良好的线性关系,酒石酸亚铁-标准曲线法适用范围广、精密性高,是一种高效、便捷的茶多酚含量测定方法,并为充分利用沙棘资源提供依据。The content of tea polyphenols in the extract of sea buckthorn tea was determined by ferrous tartrate colorimetric method,and the optimal color development conditions of the reaction were determined by single factor test and orthogonal test.The standard curve was drawn against the acid standard,and the optimal color development conditions were verified by precision test,stability test,and standard addition recovery test.The results showed that the conditions were stable,reasonable and feasible.The results showed that the optimal color development conditions were 4 mL of distilled water,5 mL of ferrous tartrate,and pH 7.5 of phosphate buffer.The concentration of tea polyphenols has a good linear relationship in the concentration range of 10-50μg/mL.The ferrous tartrate-standard curve method has a wide application range and high precision.It is an efficient and convenient method for the determination of tea polyphenols.Provide basis for making full use of sea buckthorn resources.
