摘  要:目的:通过研究糖尿病足小鼠模型给予玉米须提取物后炎症因子的变化水平,探索玉米须提取物对动物糖尿病足的影响。方法:小鼠进行适应性喂养1周,随后150只小鼠随机分成5组,空白对照组,模型对照组,玉米须提取液高、中、低剂量组,每组30只。空白对照组继续进行基础标准饲料喂养,模型对照组及玉米须提取液高、中、低剂量组给予高脂高糖饮食饲料喂养。除空白对照组外,其余4组在第12周使用STZ进行DF模型造模,开始造模前12 h小鼠禁食但可自由饮水,将STZ加入到pH值为4.2柠檬酸(浓度为0.1 mmol/L)溶解,之后的5 d按照40 mg·kg^(-1)·d^(-1)剂量给予造模小鼠行腹腔注射,造模成功标准为小鼠空腹6 h血糖值>11.1 mmol/L。将玉米须提取物按照高剂量组(400 mg/kg)、中剂量组(200 mg/kg)、低剂量组(100 mg/kg),分别配置浓度为20、10、5 g/L的治疗液,按照0.02 mL/g标准灌胃。空白及模型对照组给予等体积生理盐水,持续用药30 d,第30天给药后1 h,禁食12 h后行尾部采血进行指标检测,观察小鼠用药前后血糖、炎症因子IL-1、hs-CRP、VCAM-1、ICAM-1及MCP-1的变化。结果:与模型对照组比较,玉米须提取物高、中、低剂量组均具有降低糖尿病足小鼠模型IL-1、hs-CRP、VCAM-1、ICAM-1、MCP-1的作用(P<0.01),且呈现剂量浓度依赖性,治疗组间有差异,玉米须提取液高剂量组要明显优于低剂量组(P<0.05)。结论:玉米须提取物能够有效改善小鼠糖尿病足模型炎症因子的作用,且改善的效果随着剂量浓度的增高而增加。Objective:To study the effect of corn stigma extract on animal diabetic foot after studying the change of inflammatory factors in mice model after the intervention of corn stigma extract.Methods:150 mice were randomly divided into the blank group,the model group,and the corn stigma extract groups of high-dose,medium-dose and low-dose,with 30 mice in each group.Diabetic foot model was established by high-fat and high-sugar diet,combined with STZ injection(40 mg·kg^(-1)·d^(-1))in 12th week.The success standard of modeling was that the value of 6h fasting blood glucose was higher than 11.1 mmol/L.Corn stigma extract solution(0.02 ml/g)was applied to the high-dose group,medium-dose group and low-dose group at the concentration of 400 mg/kg(20 g/L),200 mg/kg(10 g/L)and 100 mg/kg(5 g/L),respectively.After 30 days of continuous administration,the changes of blood glucose,inflammatory factors of IL-1,hs-CRP,VCAM-1,ICAM-1 and MCP-1 were observed before and after administration in the groups.Results:Compared with those in the model group,the levels of IL-1,hs-CRP,VCAM-1,ICAM-1 and MCP-1 were significantly decreased in the corn stigma contract groups of high-dose,medium-dose and low-dose in mice with diabetic foot(P<0.01),and showed a dose-dependent manner.There were statistical differences among the groups,the high-dose group was significantly better than the low-dose group(P<0.05).Conclusion:Corn stigma extract can effectively improve inflammatory cytokines in model mice of diabetic foot,and the improving effect is increased with the increase of dose concentration.
