摘 要:目的探讨富亮氨酸α2糖蛋白1(leucine-rich alpha-2-glycoprotein 1,LRG1)及转化生长因子beta 1(transforming growth factor-beta 1,TGF-β1)在膀胱尿路上皮癌(bladder urothelial carcinoma,BUC)组织中的表达及意义。方法采用免疫组化SP法检测LRG1、TGF-β1及Ki-67在64例BUC组织及25例癌旁组织中的表达,分析LRG1、TGF-β1在各组间表达的差异及相关性。结果LRG1在BUC组织中强阳性27例(42.19%),癌旁组织中强阳性仅2例(8.00%),两组相比差异有统计学意义(P<0.05);TGF-β1在BUC组织中阳性49例(76.56%),在癌旁组织中阳性10例(40.00%),差异有统计学意义(P<0.05);相关性分析显示,LRG1与TGF-β1表达呈正相关(r^(2)=0.6582,P<0.05);LRG1、TGF-β1表达均与BUC病理学分级、浸润深度及Ki-67增殖指数相关(P<0.05)。结论LRG1、TGF-β1在BUC中表达均明显增高,两者表达均与肿瘤病理分级、浸润深度、细胞增殖指数密切相关,两者可能存在上下游调节关系。Purpose To investigate the expression and significance of leucine-rich alpha-2-glycoprotein 1(LRG1)and transforming growth factor-beta 1(TGF-β1)in bladder urothelial carcinoma.Methods Immunohistochemical SP assay was used to detect the expression of LRG1,TGF-β1 and Ki-67 in 64 cases of bladder urothelial carcinoma(BUC group)and 25 cases of paracancer tissues(control group).The difference and correlation between LRG1 and TGF-β1 expression among each group was statistically analyzed.Results The expression of LRG1 was significantly enhanced in 27 cases(42.19%)of BUC tissue,and positive in only 2 cases(8.00%)of control group.The difference between the two groups was statistically significant(P<0.05).The expression of TGF-β1 was increased in 49 cases(76.56%)of BUC tissues,and positive in 10 cases(40.00%)of control group.The difference was statistically significant(P<0.05).Correlation analysis showed that there was a clear correlation between LRG1 and TGF-β1(r2=0.6582,P<0.05).The expression of LRG1 and TGF-β1 was correlated with histopathologic grade,infiltration depth,and Ki-67 index of urothelial carcinoma(P<0.05).Conclusion The expression of LRG1 and TGF-β1 in bladder urothelial carcinoma is significantly increased,which is closely related to the pathological grade,tumor infiltration depth and cell proliferation index,and there is a positive correlation between the two proteins,which may be regulated by upstream or downstream.
