摘  要:黄芩属多年生草本植物,作为一种中药始载于《神农本草经》,是治疗感冒、流感、发烧等的常用药物。本研究以斑马鱼(5 days post fertilization)为对象,研究了不同浓度不同孵育时间对斑马鱼耐受性的影响,并在此基础上系统研究评价不同浓度黄芩对高糖诱导型(过食)和胰岛损伤型(糖尿病)斑马鱼的靶向作用功效。结果发现,斑马鱼对高浓度的黄芩具有不耐受性,孵育48 h 时的LC;为4621.832 μg/mL,95%可信范围为4031.335 ~ 5376.152μg/mL,按10% ~ 25% LC;剂量浓度推荐可用于靶向功效评价的适宜剂量浓度为462.18 ~ 1155.46 μg/mL。在10 mg/mL 葡萄糖诱导斑马鱼模型中,200 μg/mL、400 μg/mL和800 μg/mL 黄芩处理斑马鱼葡萄糖值分别降低28.32%(p > 0.05)、27.11%(p < 0.05)和35.37%(p < 0.05);在10 mg/mL 葡萄糖和0.025 mM 四氧嘧啶诱导的糖尿病斑马鱼模型中,200 μg/mL、400 μg/mL和800 μg/mL 黄芩处理组斑马鱼葡萄糖值分别降低了7.37%、8.36%和8.31%,但差异均不显著(p > 0.05)。试验表明黄芩对过饮过食与胰岛损伤型患者均具有降糖作用,其作为我国传统中药材在功能食品上具有广阔的应用前景,其叶作为保健茶饮等开发利用具有潜在价值。Scutellaria barbara, as a traditional Chinese medicine, it was first described in the Shennong Materia Medica Classic. Scutellaria barbara were commonly used to treat some diseases, like colds, flu and fever. Zebrafish(5 days post fertilization) were chosen, and we investigated the effects of different concentrations and incubation time of Scutellaria barbara on the tolerance of zebrafish in this study. On this basis, we evaluated the targeted hypoglycemic effects of different concentrations of Scutellaria barbara on high glucose-induced(overfeeding) and islet injury-(diabetes) zebrafish. The results showed that zebrafish had no tolerance to high concentration of Scutellaria barbara, and the 50% lethal concentration at 48 h incubation was 4621.832 μg/mL, and the 95% confidence range was 4031.335 to 5376.152 μg/mL. According to 10% ~ 25% of 50% lethal concentration, 462.18-1155.46 μg/mL were recommended for functional assessment. In the zebrafish model induced by 10 mg/mL glucose, the glucose value in 200, 400 and 800 μg/mL Scutellaria barbara treatments decreased by 28.32%(P > 0.05), 27.11%(P < 0.05) and 35.37%(P < 0.05), respectively. In the diabetic zebrafish model induced by 10 mg/mL glucose and 0.025 mM alloxan, the values of glucose in 200, 400 and 800 μg/mL Scutellaria barbara treatments were decreased by 7.37%, 8.36% and 8.31%, respectively, but the differences were not significant(P > 0.05). The results showed that Scutellaria barbara had a hypoglycemic effect on patients with over-eating and islet injury patients. As a traditional Chinese medicine, Scutellaria barbara has a broad application prospect for developing functional foods.
