摘  要:目的 探讨环状RNA(circRNA)circ_0014130在缺氧诱导肺腺癌侵袭转移中的作用机制。方法 收集2018年1月至2021年4月见未出现转移的肺腺癌患者46例、出现转移的肺腺癌患者42例和健康者50例。通过RT-qPCR检测缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)mRNA和circ_0014130的水平。通过Pearson检测二者的相关性。肺腺癌细胞分为对照组、缺氧组、si-circ_0014130组、缺氧+si-circ_0014130组。通过转染抑制circ_0014130,通过降低氧气浓度(10%)构建体外缺氧环境。比较各组细胞的迁移和侵袭能力。结果 肺腺癌组患者血清中的HIF-1α mRNA(2.14±0.27)和circ_0014130(2.59±0.23)水平均显著高于健康组(分别为1.00±0.16、1.00±0.14)(P<0.01);肺腺癌转移组的HIF-1α mRNA(3.85±0.42)和circ_0014130(4.36±0.44)水平均显著高于健康组和肺腺癌组(P<0.01);HIF-1α mRNA和circ_0014130的表达水平正相关(P<0.01);缺氧组的circ_0014130水平、增殖、迁移和侵袭能力显著高于对照组(P<0.01);si-circ_0014130组的circ_0014130水平、增殖、迁移和侵袭能力显著低于对照组(P<0.01);缺氧+si-circ_0014130组的circ_0014130水平、增殖、迁移和侵袭能力显著低于缺氧组且显著高于si-circ_0014130组(P<0.05)。结论 缺氧可能通过促进circ_0014130促进肺腺癌的转移。Objective To explore the mechanism of circular RNA(circRNA)circ_0014130 in hypoxia-induced invasion and metastasis of lung adenocarcinoma.Methods All 46 patients with no metastases,42 patients with metastases,and 50 healthy patients were collected between January 2018 and April 2021. The levels of anoxically induced factors-1α(hypoxiainducible factor-1α,HIF-1α)m RNA and circ_0014130 were detected by RT-qPCR. The correlation was detected by Pearson. Pulmonary adenocarcinoma cells were divided into control group,hypoxia group,si-circ_0014130 group,hypoxic + si-circ_0014130 group. Inbits circ_0014130,by transfection by reducing oxygen concentration(10%). Compare the migration and invasion ability of the cells in each group.Results The serum levels of HIF-1α mRNA(2.14±0.27)and circ_0014130(2.59±0.23)in the lung adenocarcinoma group were significantly higher than those in the healthy group(1.00±0.16;1.00±0.14)(P<0.01). The levels of HIF-1α mRNA(3.85±0.42)and circ_0014130(4.36±0.44)in the lung adenocarcinoma metastasis group were significantly higher than those in the healthy group and lung adenocarcinoma group(P<0.01). HIF-1αmRNA was positively correlated with circ_0014130(P<0.01). The circ_0014130 level,proliferation,migration and invasion abilities of the hypoxia group were significantly higher than those of the control group(P<0.01). The circ_0014130 level,proliferation,migration and invasion abilities of the si-circ_0014130 group were significantly lower than those of the control group(P<0.01). The level of circ_0014130,proliferation,migration and invasion abilities of hypoxia + si-circ_0014130group were significantly lower than those of hypoxia group and significantly higher than those of si-circ_0014130 group(P<0.01).Conclusion Hypoxia may promote the metastasis of lung adenocarcinoma by promoting circ_0014130.
