摘  要:目的 研究烟草烟雾提取物(CSE)对人牙龈成纤维细胞(HGF)增值的影响。方法 采用组织块法原代培养HGF并鉴定其来源,制备含有不同浓度CSE培养液,采用四唑盐(MTT)比色法测定不同浓度烟草烟雾对HGF增殖的影响。结果 作用24、48、72 h时,空白对照的光密度(OD)值分别为(0.2812±0.0296)、(0.3894±0.0298)、(0.5059±0.0269), 2.5%CSE的OD值分别为(0.1832±0.0342)、(0.2959±0.0214)、(0.3153±0.0219), 5.0%CSE的OD值分别为(0.1379±0.0298)、(0.1423±0.0134)、(0.1586±0.0379), 10.0%CSE的OD值分别为(0.1060±0.0246)、(0.1031±0.0169)、(0.0920±0.0161), 20.0%CSE的OD值分别为(0.0861±0.0142)、(0.0789±0.0190)、(0.0726±0.0159)。空白对照、2.5%CSE、5.0%CSE、10.0%CSE、20.0%CSE的OD值均逐渐降低,且2.5%CSE、5.0%CSE、10.0%CSE、20.0%CSE的OD值均低于空白对照,差异具有统计学意义(P<0.05)。结论 烟草烟雾可抑制HGF的增殖,并随着培养液浓度的增大,抑制作用愈来越明显,具有浓度依赖性。Objective To study the effect of cigarette smoke extract(CSE)on the proliferation of human gingival fibroblasts(HGF).Methods Primary tissue mass method was used to culture human gingival fibroblasts and identify their source.The culture medium containing different concentrations of CSE was prepared,and the effect of different concentrations of cigarette smoke on the proliferation of HGF was determined by methyl thiazolyl tetrazolium(MTT)colorimetric method.Results The optical density(OD)values of the blank control at 24,48 and 72 h of action were(0.2812±0.0296),(0.3894±0.0298),(0.5059±0.0269),the OD values for 2.5%CSE were(0.1832±0.0342),(0.2959±0.0214),(0.3153±0.0219),the OD values for 5.0%CSE were(0.1379±0.0298),(0.1423±0.0134),(0.1586±0.0379),the OD values for 10.0%CSE were(0.1060±0.0246),(0.1031±0.0169),(0.0920±0.0161),and the OD values for 20.0%CSE were(0.0861±0.0142),(0.0789±0.0190),and(0.0726±0.0159).The OD values of the blank control,2.5%CSE,5.0%CSE,10.0%CSE,and 20.0%CSE gradually decreased,and the OD values of 2.5%CSE,5.0%CSE,10.0%CSE,and 20.0%CSE were all lower than those of the blank control.All the differences were statistically significant(P<0.05).Conclusion Cigarette smoke can inhibit the proliferation of HGF,and the inhibitory effect becomes more and more obvious with the increase of the concentration of culture medium,which has a concentration dependence.
