摘 要:目的探究银杏叶提取物通过自由基清除作用保护脑缺血大鼠钠、钾-三磷酸腺苷酶(Na,K-ATPase)活性。方法选取40只成年大鼠,均进行大脑中动脉闭塞手术,建立脑缺血模型,排除死亡个体4例,使用随机数字表法分为4组(对照组,低剂量组,中剂量组,高剂量组)分别给予安慰剂及不同剂量银杏叶提取物,干预48h后处死大鼠,取脑皮质,观察各组脑皮质病理变化并比较各组Na,K-ATPase活性。检测各组大鼠MDA、SOD、CAT及炎症因子水平并观察其TLR4、NF-κB蛋白表达。结果对照组大鼠脑皮质结构受损严重,表现为细胞皱缩、空泡变性、细胞着色不均、排列紊乱,神经细胞凋亡,研究组大鼠脑皮质病理变化明显减轻,且随银杏叶提取物剂量增加进一步缓解;对照组Na,K-ATPase活性显著低于低剂量组、中剂量组、高剂量组,且随剂量上升,其活性均呈现升高趋势;对照组MDA水平显著高于低剂量组、中剂量组、高剂量组,且随剂量上升,MDA呈现降低趋势(P<0.05);对照组SOD、CAT水平显著低于低剂量组、中剂量组、高剂量组,且随剂量上升,呈现升高趋势(P<0.05);对照组IL-1β、IL-6、TNF-α水平显著高于低剂量组、中剂量组、高剂量组,且随剂量上升,其水平均呈现降低趋势(P<0.05);对照组TLR4、NF-κB蛋白及mRNA相对表达量显著高于低剂量组、中剂量组、高剂量组,且随剂量上升,其水平均呈现降低趋势(P<0.05)。结论银杏叶提取物可通过调控脑缺血大鼠TLR4/NF-κB通路及自由基清除作用保护脑缺血大鼠Na,K-ATPase活性,从而抑制其大脑炎性因子释放及神经元细胞凋亡,保护脑缺血大鼠脑组织。Objective To explore the protection of Ginkgo biloba extract through free radical scavenging effect on sodium and potassium-adenosine triphosphate(Na,K-ATPase)activity in rats with cerebral ischemia.Methods 40 adult rats were selected as the research sample,and all of them underwent middle cerebral artery occlusion surgery with established cerebral ischemia model.We excluded 4 dead subjects,and divided them into 4 groups using random number table method(control group,low-dose group,middle-dose group and high-dose group),which were given placebo and different doses of Ginkgo biloba extract.After 48 hours of intervention,these rats were sacrificed and the cerebral cortex was taken to observe pathological changes of the cerebral cortex in each group and compare Na,K-ATPase activities of each group.We detected levels of MDA,SOD,CAT and inflammatory factors in each group of rats and observed their TLR4 and NF-κB protein expressions.Results The cerebral cortex structure of the rats in the control group was severely damaged,manifested as cell shrinkage,vacuolar degeneration,uneven cell coloration,disordered arrangement,and neuronal apoptosis.Pathological changes of the cerebral cortex of the rats in the study group were significantly reduced.The increase in the dose of leaf extract was further relieved;Na,K-ATPase activities of the control group were significantly lower than those of the low-dose,middle-dose,and high-dose groups,and their activities showed an increasing trend as the dose increased;MDA level of the control group was significantly higher than that of the low-dose,medium-dose,and high-dose groups,and with the increase in the dose,MDA showed a decreasing trend(P<0.05);The control group had significantly lower levels of SOD and CAT compared with the low-dose group,middle-dose group,and high-dose group.MDA showed an increasing trend with increasing dose(P<0.05);IL-1β,IL-6,and TNF-αlevels in the control group were significantly higher than those in the low-dose group,the middle-dose group and high-dose grou
