摘 要:为深入研究牛乳和驼乳中酪蛋白的差异蛋白组学分析,采用双向电泳和质谱联用技术在蛋白质水平上探讨牛乳和驼乳的差异性,获得其中差异蛋白质,并以此为标识物建立了驼乳中牛源性成分的检测方法,进一步促进驼乳以及驼乳酪蛋白的研究和开发。用双向电泳技术(two-dimensional gel electrophoresis,2-DE)对牛乳和驼乳中酪蛋白进行了初步分离分析。结果表明:与牛乳酪蛋白相比,在驼乳酪蛋白的2-DE图谱中发现差异蛋白斑点有40个,其中33个蛋白在驼乳酪蛋白中表达缺失,7个蛋白在驼乳酪蛋白中表现为低表达,对低表达的蛋白斑点(10、11、13、16、63、64、127)进行Maldi-TOF-TOF质谱鉴定,成功回收和鉴定了这7个低表达蛋白的中5个,归属为3个蛋白,分别是酪蛋白2、κ-酪蛋白A片段、β-酪蛋白。说明驼乳和牛乳酪蛋白之间的确实存在一定差异性,可以以此差异蛋白为检测标识物来建立驼乳中牛源性成分的检测方法,来满足驼乳及乳制品真实性问题的检测需求。In order to investigate the differential proteomic analysis of casein in bovine and camel milk in depth, this study used bi-directional electrophoresis and mass spectrometry to explore the differences between bovine and camel milk at the protein level, to obtain the differential proteins among them, and to use them as markers to establish a method for the detection of bovine-derived components in camel milk, and to further promote the research and development of camel milk as well as camel milk casein.In this paper, two-dimensional gel electrophoresis(2-DE) was used to initially analyze the differential proteins with matching protein spots in bovine and camel milk casein. The results showed that 40 differential protein spots were identified in the 2-DE profiles of camel milk casein compared to bovine milk casein, of which 33 proteins were missing in camel milk casein and 7 proteins showed low expression in camel milk casein, and the low-expressed protein spots(10,11, 13, 16, 63, 64, 127) were identified by Maldi-TOF-TOF mass spectrometry. Five of these seven low-expressed proteins were successfully recovered and identified, attributed to three proteins, casein 2, κ-casein A fragment, and β-casein, respectively.It is suggested that there are some differences between camel milk and bovine milk casein, and the differential protein can be used as a marker to establish a method for the detection of bovine-derived components in camel milk to meet the demand of authenticity of camel milk and dairy products.
