摘  要:目的 以高脂饲料喂养的小鼠为高脂模型,研究黑茶叶提取物的预防性降血脂作用及药效物质基础。方法 以高、中、低不同剂量黑茶水提取物灌胃给药5周,期间每隔5d记录各组小鼠进食量、饮水量及体重变化;实验结束后,测定各组小鼠血清中的总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)水平;同时,利用超高效液相色谱-高分辨飞行时间质谱联用(UPLC-QTOF-MS/MS)技术对黑茶叶的主要化学成分进行初步研究。结果 与高脂模型组小鼠相比,黑茶水提取物可以显著预防由高脂饲料诱发的血清脂质代谢异常,包括呈剂量依赖性地降低高脂小鼠血清中TC、LDL-C的水平,降低了TC/HDL-C和AI指数。UPLC-QTOF-MS/MS分析表明黑茶叶中的化学组成主要包括原花色素及其衍生物类成分。结论 黑茶水提取物具有明显地预防改善高脂小鼠脂质代谢紊乱的作用,其药效物质基础成分可能主要包括原花色素、黄酮及其衍生物类化合物。Objective To study the preventive effect of dark tea on high-fat diet-induced hyperlipidemic mice and its bioactive components. Methods The water extract of dark tea in different doses gavage groups of hyperlipidemic mice during the 5 weeks of experiment. Food,water intakes and body weight were check every 5 days. The effects of dark tea on serum levels of total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDLC)were investigated. Meanwhile,the major bioactive components of dark tea were analyzed by ultra-high performance liquid chromatography-quadrupole time of flight tandem mass spectrometry(UPLC-QTOF-MS/MS). Results The serum TC,LDLC were significantly decreased,and the serum TC/HDL-C ratio and AI index were obviously increased compared with model group of hyperlipidemic mice in a dose-dependent manner. Meanwhile,the major bioactive components of dark tea were identified as proanthocyanidins and their derivatives by UPLC-QTOF-MS/MS. Conclusion The dark tea possessed obvious preventive effect on dyslipidemias induced by high-fat diet,and its major bioactive components were identified as proanthocyanidins,flavonoids and their derivatives.
