摘 要:THP-1细胞经LPS和IFN-γ活化诱导为经典激活的M1型巨噬细胞。M1型巨噬细胞在肿瘤发生的初期发挥着重要的作用,能够识别并清除肿瘤细胞,但对食管鳞癌细胞Eca109的具体调节作用尚不清楚。本研究利用THP-1细胞来源的M1型巨噬细胞与食管鳞癌细胞Eca109共培养的模型;用实时荧光定量聚合酶链式反应(qRT-PCR)检测TNF-α和IL-1β mRNA的表达;用流式细胞技术、Transwell实验检测M1型巨噬细胞与食管鳞癌细胞Eca109共培养24 h后对Eca109细胞凋亡、增殖与迁移的影响。结果显示,诱导后的THP-1细胞完全贴壁并有伪足出现;与M0细胞相比,诱导的M1型巨噬细胞相关细胞因子TNF-α、IL-1β的mRNA表达均显著增高(P<0.01);与M1型巨噬细胞共培养后可以促进食管鳞癌细胞的凋亡(P<0.01)、抑制食管鳞癌细胞的增殖(P<0.01)与迁移(P<0.001)。本研究结果提示,THP-1来源的M1型巨噬细胞与食管鳞癌细胞Eca109共培养可以促进食管鳞癌细胞Eca109的凋亡,抑制其增殖与迁移。THP-1 cells are induced into classic activated M1 macrophages by LPS and IFN-γactivation.M1macrophages play an important role in the early stage of tumorigenesis.M1 macrophages can recognize and remove tumor cells,but the specific regulatory effect on esophageal squamous cell carcinoma cell line Eca109is not clear.In this study,the model of co-culture of M1 macrophages derived from THP-1 cells with esophageal squamous cell carcinoma cell line Eca109 was used,the expression of TNF-αand IL-1βwas detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR),and the effect of M1macrophage co-cultured with esophageal squamous cell carcinoma cell line Eca109 on apoptosis,proliferation and migration of Eca109 cells were detected by flow cytometry and transwell assay.The results showed that the induced THP-1 cells adhered to the wall completely and had pseudopodia,and the mRNA expression of TNF-αand IL-1βof M1 macrophages was significantly higher than that of M0 cells(P<0.01).Co-culture with M1 macrophages could promote apoptosis and inhibit the proliferation and migration of esophageal squamous cell carcinoma cells(P<0.01).The results suggest that co-culture of THP-1-derived M1 macrophages with esophageal squamous cell carcinoma cell line Eca109 can promote the apoptosis(P<0.01)of esophageal squamous cell carcinoma cell line Eca109 and inhibit its proliferation(P<0.01)and migration(P<0.001).
