摘 要:目的:研究白细胞介素-24(interleukin 24,IL-24)通过Janus相关激酶2(Janus kinase 2,JAK2)/信号转导与转录激活因子3(signal transduction and activator of transcription 3,STAT3)途径调控口腔鳞癌细胞增殖的作用及机制。方法:培养口腔鳞癌细胞株HN4、HN6、HN30及人口腔黏膜角质细胞株HOK,检测IL-24的表达量;筛选出IL-24低表达的HN30细胞并分组,分别转染空白的GV608质粒、表达IL-24的重组GV608质粒或加入IL-6处理,MTS法检测细胞增殖活力。Western blot检测细胞中IL-24、B淋巴细胞瘤-2基因(bcl-2)、细胞周期蛋白D1(cyclinD1)、血管内皮生长因子(VEGF)、p-JAK2、p-STAT3的表达量。结果:HN4、HN6、HN30细胞中IL-24的表达均低于HOK细胞(P<0.05),且HN30细胞中IL-24表达减少最显著;IL-24质粒组HN30细胞增殖活力的OD_(490nm)水平及细胞中bcl-2、cyclinD1、VEGF、p-JAK2、p-STAT3的表达量均低于阴性对照组、空白质粒组(P<0.05)。IL-24质粒+IL-6组HN30细胞增殖活力的OD_(490nm)水平及细胞中bcl-2、cyclinD1、VEGF的表达量均明显高于IL-24质粒组(P<0.05)。结论:过表达IL-24对口腔鳞癌细胞株HN30的增殖具有抑制作用且该作用与抑制JAK2/STAT3途径有关。Objective:To study the effect and mechanism of of interleukin-24(IL-24)on regulating the proliferation of oral squamous cell carcinoma cell via regulating Janus kinase 2(JAK2)/signal transduction and activator of transcription 3(STAT3)pathway.Methods:Human oral squamous cell carcinoma cell lines HN4,HN6,HN30 and human oral keratinocyte line HOK were cultured and the expression of IL-24 was detected.The low expression of IL-24 cell line HN30 was screened and divided into groups.Then cells were transfected with blank GV608 plasmid,the recombinant GV608 plasmid expressing IL-24 or treated with IL-6.Cell proliferation was detected by MTS,the expression of IL-24,B lymphoma-2 gene(bcl-2),cyclinD1,VEGF,p-JAK2 and p-STAT3 were detected by Western blot.Results:The expression of IL-24 in HN4,HN6,HN30 cells was lower than that in HOK cells(P<0.05),and the expression of IL-24 in HN30 cells decreased most significantly.The OD_(490 nm) level of proliferation activity and the expression of bcl-2,cyclinD1,VEGF,p-JAK2,p-STAT3 in HN30 cells of IL-24 plasmid group were lower than those of negative control group and blank plasmid group(P<0.05).The OD_(490 nm) level of proliferation activity and the expression of bcl-2,cyclinD1,VEGF,p-JAK2,p-STAT3 in HN30 cells of IL-24 plasmid+IL-6 plasmid group were higher than those in IL-24 plasmid group(P<0.05).Conclusion:Over expression of IL-24 inhibits the proliferation of oral squamous cell carcinoma cell line HN30,which is related to the inhibition of JAK2/STAT3 pathway.
