摘 要:目的对20株不动杆菌进行分子特征描述和比较研究,用于鲍曼不动杆菌参考品的研制。方法结合生化鉴定、16S rRNA基因序列分析、rpoB基因序列分析和基质辅助激光解吸电离飞行时间质谱等四种鉴定方法对菌株进行精准鉴定,并通过脉冲场凝胶电泳(pulsed field gel electrophoresis,PFGE)和多位点序列分型(multilocus sequence typing,MLST)进行多样性和进化关系的分析。结果综合比较不同方法的鉴定结果,完成20株不动杆菌的精准鉴定。分子分型显示,菌株具有多样性,且进化关系清晰,发现了16种新基因型和7种新ST型。结论本研究通过组合鉴定方式精准鉴定参考品制备用候选不动杆菌,并分析了菌株的多样性和进化关系,为参考品研制提供资源支持。Objective To perform molecular characterization and comparative study of 20 strains of Acinetobacter spp.for the development of reference materials of Acinetobacter baumannii.Methods The strains were precisely identified by a combination of biochemical identification,16 S rRNA gene sequence analysis,rpoB gene sequence analysis and matrix-assisted laser desorption ionization time-of-flight mass spectrometry,and analyzed for diversity and evolutionary relationships by pulsed field gel electrophoresis(PFGE)and multilocus sequence typing(MLST).Results The precise identification of 20 strains was completed through the comprehensive comparison of the identification results of different methods.Molecular typing showed that the strains were diverse and had clearly evolutionary relationships.Meanwhile,16 new genotypes and 7 new ST types were identified.Conclusion This study accurately identified the candidate strains for the preparation of reference materials through combined identification methods,and analyzed the diversity and evolutionary relationship among the strains,which provide resource to support the development of reference materials.
