摘 要:目的:探讨七氟醚对胃癌HGC-27细胞增殖、迁移和侵袭的影响及作用机制。方法:不同浓度的七氟醚作用于HGC-27细胞后,四甲基噻唑蓝染色法(MTT)检测细胞增殖,Transwell检测细胞的迁移和侵袭,Western blot检测细胞中细胞周期蛋白D1(CyclinD1)、细胞核增殖相关抗原(Ki67)、基质金属蛋白酶(MMP)-2和MMP-9蛋白水平,实时荧光定量PCR(RT-qPCR)检测细胞中lncRNA ASBEL水平。转染ASBEL的小干扰RNA至HGC-27细胞中,或转染ASBEL的过表达载体pcDNA3.0-ASBEL至HGC-27细胞中并以4%七氟醚作用细胞,采用上述相同方法观察沉默ASBEL表达对HGC-27细胞增殖、迁移和侵袭及相关蛋白CyclinD1、Ki67、MMP-2和MMP-9蛋白表达的影响。结果:七氟醚可降低HGC-27细胞存活率、迁移和侵袭细胞数及CyclinD1、Ki67、MMP-2和MMP-9蛋白表达(P<0.05),降低HGC-27细胞ASBEL基因表达(P<0.05)。沉默ASBEL表达可降低HGC-27细胞存活率、迁移和侵袭细胞数及CyclinD1、Ki67、MMP-2和MMP-9蛋白表达(P<0.05)。过表达ASBEL逆转了七氟醚对胃癌HGC-27细胞存活率、迁移和侵袭及CyclinD1、Ki67、MMP-2和MMP-9蛋白表达的抑制作用(P<0.05)。结论:七氟醚可能通过下调ASBEL表达抑制胃癌HGC-27细胞的增殖、迁移和侵袭。Objective:To investigate the effects and mechanism of sevoflurane on proliferation,migration and invasion of gastric cancer HGC-27 cells and its mechanism.Methods:After different concentrations of sevoflurane were applied to HGC-27 cells,cell proliferation was detected by tetramethylthiazole blue staining(MTT)assay,migration and invasion of cells were detected by Transwell,the expression levels of cyclin D1(CyclinD1),Nuclear proliferation-associated antigen(Ki67),matrix metalloproteinase(MMP)-2 and MMP-9 protein levels were detected by Western blot,and lncRNA ASBEL levels in cells was detected by RT-qPCR.After transfecting small interfering RNA of ASBEL into HGC-27 cells,or transfecting ASBEL overexpression vector pcDNA3.0-ASBEL into HGC-27 cells and treated with 4%sevoflurane,the effects of silencing ASBEL expression on the proliferation,migration and invasion of HGC-27 cells and the expression of related proteins CyclinD1,Ki67,MMP-2 and MMP-9 were observed.Results:Sevoflurane reduced the survival rate,migration and invasion of HGC-27 cells and the expression levels of CyclinD1,Ki67,MMP-2 and MMP-9 protein(P<0.05),and decreased the expression level of ASBEL gene in HGC-27 cells(P<0.05).Silencing ASBEL expression reduced HGC-27 cell survival rate,migration and invasion number,and the expression levels of CyclinD1,Ki67,MMP-2 and MMP-9 protein(P<0.05).Overexpression of ASBEL reversed the inhibitory effect of sevoflurane on the survival,migration and invasion of gastric cancer HGC-27 cells and the expression of CyclinD1,Ki67,MMP-2 and MMP-9 proteins(P<0.05).Conclusion:Sevoflurane may inhibit the proliferation,migration and invasion of gastric cancer HGC-27 cells by down-regulating ASBEL expression.
