摘  要:目的探讨血管生成素-1(Ang-1)、血管生成素-2(Ang-2)及受体Tie-2在妊娠滋养细胞疾病中的表达变化及意义。方法运用实时荧光定量RT-PCR技术分别检测18例部分性葡萄胎、19例完全性葡萄胎、15例侵蚀性葡萄胎、20例绒毛膜癌和20例正常妊娠者绒毛组织中Ang-1、Ang-2、Tie-2 mRNA的表达,Western blot印迹法检测组织中Ang-1、Ang-2、Tie-2蛋白质的表达。结果正常妊娠组相比,部分性葡萄胎组、完全性葡萄胎组、侵蚀性葡萄胎组、绒毛膜癌组Ang-1、Ang-2、Tie-2 mRNA及其蛋白质的表达水平依次显著增高,绒毛膜癌组最高,差异均有统计学意义(P<0.05)。结论滋养细胞Ang-1、Ang-2及其受体Tie-2表达增高,可能在妊娠滋养细胞疾病的发病机制中发挥重要作用。Objective To explore the changes of expressions of angiogenin-1(Ang-1),angiogenin-2(Ang-2),and the receptor(Tie-2)in gestational trophoblastic diseases and the significance.Methods Real-time fluorescence quantitative PCR was used to detect the expressions of Ang-1,Ang-2,and Tie-2 mRNA in villi tissue of 18 cases of partial hydatidiform mole,9 cases of complete hydatidiform mole,15 cases of invasive hydatidiform mole,20 cases of choriocarcinoma,and 20 normal pregnant women.Western blot was used to detect the expressions of Ang-1,Ang-2,and Tie-2 proteins.Results Compared with normal pregnancy group,the expression levels of Ang-1,Ang-2,Tie-2 mRNA and proteins increased gradually in partial hydatidiform mole group,complete hydatidiform mole group,invasive hydatidiform mole group,and choriocarcinoma group,the levels in choriocarcinoma group were the highest,there were statistically significant differences(P<0.05).Conclusion The expression levels of Ang-1,Ang-2,and Tie-2 in trophocytes increase,which may play important roles in pathogenesis of gestational trophoblastic diseases.
