摘 要:目的多糖是石斛主要生物活性成分之一。本文以8种常见石斛为材料,对其多糖相关特征和生物活性进行比较研究,提高石斛多糖在制药、日常保健、美容等加工领域的深度与精度,有利于其进一步发展。方法以苯酚-硫酸法检测石斛多糖的含量,以凝胶渗透色谱法分析石斛多糖的组分,并测定其非细胞抗氧化活性。结果多糖含量在所测的8种石斛里差异明显,铁皮石斛的多糖含有量为43.83%,球花石斛多糖含有量仅为15.93%。8种石斛多糖均为非单一组分,组分最多的是鼓槌石斛(5个),最少的是美花石斛(2个)。8种石斛多糖的分子量大小差异较大,范围在3.62×1012~1.04×1015Da。不同种类的石斛其多糖在抗氧化性方面也具有不同的效果,且具有较大差异,对于抑制脂质的过氧化而言,能力最强的是束花石斛多糖,达到33.23%,而细茎石斛的还原力为0.20,是几种石斛中的最佳,清除DPPH游离基的能力也是最高的27.43%,其余石斛的抗氧化活性也在不同方面有所体现。结论石斛多糖的含量、组成和生物学活性在种间差异明显。虽然铁皮石斛多糖含量最高,鼓槌石斛多糖组分最复杂,但是抗氧化活性各种石斛各有优势。OBJECTIVE To explore the characteristics of eight kinds of Dendrobium species polysaccharides and and compare their biological activities.METHODS The content of Dendrobium polysaccharides were detected by phenol-sulfuric acid method,the components of Dendrobium polysaccharides were analyzed by gel permeation chromatography,and their acellular antioxidant activities were determined.RESULTS The polysaccharide contents of the eight Dendrobium species were significantly different.The polysaccharide content of D.officinale were 43.83%,and the polysaccharide content of D.thyrsiflorum was only 15.93%.The components of polysaccharides from eight Dendrobium species were nonsingle fraction.The number of components of D.chrysotoxum was the most(5 fractions)but D.loddigesii were the least(2 fractions).The molecular weight distribution of eight Dendrobium species polysaccharides were quite different,ranging from 3.62×10^(12)-1.04×10^(15)Da.Different types of Dendrobium polysaccharides also had different effects on antioxidant properties and showed great differences.D.chrysanthum polysaccharide was the best effect on the inhibition of lipid peroxidation,reaching 33.23%.The reducing power and DPPH free radical scavenging capacity of D.moniliforme were both best,reaching 0.20 and 27.43%.The antioxidant activities of other Dendrobium species were also reflected in different aspects.CONCLUSION There are great differences in the contents,composititions and biological activities of polysaccharides among different Dendrobium species.Although D.officinale has the highest polysaccharide content and D.chrysotoxum has the most complex polysaccharide components,all of Dendrobium have their own advantages on antioxidant activities.
