摘 要:目的 研究人脐带源间充质干细胞(hUC-MSCs)移植对CCl_(4)所致肝硬化大鼠的治疗及抗氧化作用。方法 皮下注射CCl_(4)橄榄油溶液制备大鼠肝硬化模型,造模成功后尾静脉注射hUC-MSCs,并于hUC-MSCs移植后第8周处死大鼠并检测大鼠血清谷氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、总胆红素(TBIL)、直接胆红素(DBIL)和清蛋白(ALB)等肝功能指标,以及超氧化物歧化酶(SOD)、谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GPx)和丙二醛(MDA)等氧化应激指标,应用苏木精-伊红染色和MASSON三色染色评估肝细胞炎性浸润及纤维沉积。结果 hUC-MSCs移植可以改善CCl_(4)所致肝硬化大鼠的肝功能,改善大鼠切片病理学炎症评分以及纤维组织沉积;hUC-MSCs移植可升高CCl_(4)所致肝硬化大鼠SOD、GSH、GPx表达,降低MDA表达。结论 hUC-MSCs移植可改善CCl_(4)所致肝硬化大鼠的肝功能,改善肝硬化大鼠肝细胞炎性浸润及纤维化沉积,抗氧化作用可能是其作用机制。Objective To study the therapeutic and antioxidant effects of human umbilical cord derived mesenchymal stem cells(hUC-MSCs)transplantation on CCl_(4) induced liver cirrhosis in rats.Methods Wistar rats were induced by CCl_(4) to make liver cirrhosis model.After successful modeling,hUC-MSCs were injected into caudal vein,and the rats were killed eight weeks after hUC-MSCs transplantation.The liver function indexes such as serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),direct bilirubin(DBIL)and albumin(ALB)were measured,as well as superoxide dismutase(SOD),glutathione(GSH),glutathione peroxidase(GPx)and malondialdehyde(MDA)and other oxidative stress indicators.Hepatocyte inflammatory infiltration and fibrous deposition were assessed by hematoxylin-eosin staining and MASSON trichrome staining.Results hUC-MSCs transplantation could improve the liver function,pathological inflammation score and fibrous tissue deposition in CCl_(4) induced cirrhotic rats.hUC-MSCs transplantation could increase the expression of SOD,GSH and GPX and reduce the expression of MDA in cirrhotic rats induced by CCl_(4).Conclusions hUC-MSCs transplantation has a therapeutic effect on CCl_(4) induced liver cirrhosis in rats,by improving the liver function,inflammatory infiltration and fibrotic deposition of liver cells.Antioxidation may be its mechanism.
