摘 要:[目的]优化野蘑菇三萜类化合物的最佳提取工艺。[方法]以野蘑菇菌种培养所得的菌丝体为原料,采用超声波辅助、异丙醇-硫酸铵双水相体系提取野蘑菇中的三萜类化合物,并建立检测野蘑菇中三萜类化合物的紫外分光光度法(UV法)。对影响三萜类化合物提取率的主要因素--异丙醇体积浓度、硫酸铵质量浓度、提取时间进行单因素试验,在此基础上根据Box-Benhnken试验设计原理利用响应面法对提取工艺进行优化。[结果]野蘑菇菌丝体内三萜类化合物的提取最佳工艺条件为异丙醇体积浓度45%,硫酸铵浓度0.175 g/mL,超声波提取时间35 min(320 W 4℃)。在该条件下,三萜提取率为最高。[结论]该研究为野蘑菇中三萜类化合物的提取及进一步开发利用奠定了基础。[Objective]Optimization of the extraction process of triterpenoids from Agaricus arvensis Schaeff ex Fr..[Method]The triterpenoids in Agaricus arvensis Schaeff ex Fr.mycelium were extracted by ultrasonic-assisted isopropanol ammonium sulfate aqueous two-phase system,and an ultraviolet spectrophotometry(UV method)was established for the detection of the triterpenoids.Single factor experiments were carried out on the main factors affecting the extraction rate of triterpenoids:isopropanol volume fraction,ammonium sulfate mass concentration and ultrasonic-assisted extraction time.On this basis,the extraction process was optimized by response surface method according to the Box-Benhnken experimental design principle.[Result]The results showed that the optimum extraction conditions of triterpenoids from Agaricus arvensis Schaeff ex Fr.Mycelium were as follows:45%isopropyl alcohol,0.175 g/mL ammonium sulfate,ultrasonic extraction time of 35 min(320 W 4℃).Under these conditions,the extraction rate of triterpenoids was the highest.[Conclusion]This study can lay a foundation for the extraction and further utilization of triterpenoids from Agaricus arvensis Schaeff ex Fr.
