摘 要:目的:观察青蒿琥酯(Art)对牙周炎大鼠的治疗作用及可能机制。方法:60只牙周炎大鼠随机分为模型组,Art低、中、高剂量组,各15只,另15只大鼠为对照组。Art低、中、高剂量组每只大鼠分别腹腔注射Art 10、30、60 mg/kg,每周1次,连续4周。对照组、模型组腹腔注射等体积生理盐水。治疗结束后检测牙龈菌斑指数(PLI)、出血指数(SBI),牙周组织骨保护蛋白(OPG)和RANKL蛋白表达量。结果:与模型组比较,Art低剂量组PLI、SBI降低,OPG表达量升高(P<0.05),RANKL表达降低(P<0.05),且随着Art剂量的增加各指标改善更为明显。结论:Art可抑制牙周炎大鼠牙槽骨吸收,减轻炎症反应,可能通过增加OPG表达,降低RANKL表达发挥作用。Objective:To observe the effects of artesunate(Art)on alveolar bone resorption and anti-inflammatory effects in rats with periodontitis,and to study the possible mechanism.Methods:60 periodontitis rats were randomly divided into model group,Art of low,medium and high dose groups(n=15),and another 15 rats were served as the control group.Art of 10,30,and 60 mg/kg were respectively injected intraperitoneally into the rats of Art low,medium,and high dose groups,once a week for 4 weeks,and the same amount of normal saline was injected intraperitoneally in the rats of control group and the model group.After treatment,the gingival plaque index(PLI)and bleeding index(SBI)were measured,osteoprotective protein(OPG)and RANKL protein expression in periodontal tissue were detected.Results:Compared with the model group,PLI,SBI in the Art low-dose group were decreased,OPG protein expression was increased(P<0.05),RANKL expression decreased(P<0.05),and with the increase of Art dose,the improvement of each index was more obvious.Conclusion:Art can inhibit alveolar bone resorption and reduce inflammation in rats with periodontitis.It may play a role by increasing OPG protein expression and reducing RANKL protein expression.
