摘 要:目的:利用气相色谱-质谱联用(GC-MS)技术对寒热体质大鼠进行非靶向代谢组学研究,观察寒、热体质大鼠尿液代谢谱的差异性,以期找出与代谢特征有关的差异代谢物和代谢通路。方法:采用GC-MS技术对大鼠尿液进行非靶向代谢组学检测,结合主成分分析(PCA)和正交偏最小二乘判别分析(OPLS-DA)挖掘出寒、热体质之间的差异性代谢物,关联相关代谢途径,阐释造成寒热体质差异的生物学机制。结果:筛选并鉴定出热体组与常体组间有21个差异性代谢物及7条代谢通路,寒体组与常体组间有12个差异性代谢物及4条代谢通路。差异性代谢产物主要包括糖类、氨基酸、有机酸、维生素及脂肪酸等。结论:常体、寒体和热体体质大鼠尿液代谢组学存在差异,3种体质间存在不同的最终差异代谢物和差异代谢通路,主要与机体的能量代谢等有关。初步揭示了造成寒体、热体与常体差异的机制,对继续深入研究体质可调性具有重要的意义。Objective:To find out the different metabolites and metabolic pathways related to metabolic characteristics,non-targeted metabolisms was applied in research of cold and heat body constitution rats by GC-MS.Methods:GC-MS was used to detect the non-targeted metabolisms of rat urine,PCA and OPLS-DA were used to find out the different metabolites between cold and heat body constitution,and related metabolic pathways were associated to explain the biological mechanism of the difference in cold and heat body constitution.Results:A total of 21 differential metabolites and 7 metabolic pathways between heat and normal body constitution group,12 differential metabolites and 4 metabolic pathways between cold and normal body constitution group were screened and identified.Differential metabolites mainly include sugars,amino acids,organic acids,vitamins and fatty acids,and so on.Conclusion:There are significant differences in urine metabolisms among normal,cold and heat body constitution rats.There are separate final differential metabolites and differential metabolic pathways among the three body constitutions,which are mainly related to the energy metabolism of the body.This study preliminarily reveals the mechanism of the difference between normal,cold and heat body constitution,which is of great significance to further study the adjustability of constitution.
