摘 要:目的:研究乳癌术后方通过SDF-1/CXCR4信号通路对乳腺癌肺转移小鼠模型中肺转移灶的影响。方法:建立4T1乳腺癌小鼠移植瘤肺转移模型,随机分为模型组、中药组、西药组、联合用药组,观察各组小鼠肺转移情况;采用RT-PCR、Western Blot法及免疫组化法检测肺组织中SDF-1/CXCR4信号通路相关分子mRNA及蛋白表达。结果:接种4周后小鼠的成瘤率及肺转移率为100%。中药组、西药组、联合用药组的肺转移抑制率为42.85%、49.35%、52.59%,较模型组显著下降(P<0.05)。与模型组比较,各治疗组NF-κB、CXCR4、CXCL12、CD147 mRNA表达显著降低(P<0.05,P<0.01);同时CXCR4、SDF-1、NF-κB、p-NF-κB、CD147蛋白表达亦显著下降(P<0.01)(中药组CD147除外)。结论:乳癌术后方可以抑制乳腺癌肺转移发生,同时降低肺转移灶中SDF-1/CXCR4信号通路相关分子的表达。Objective:To study the effect of Ru’ai Shuhou Formula on lung metastases in a mouse model of breast cancer lung metastasis through SDF-1/CXCR4 signaling pathway.Methods:4T1 breast cancer mouse transplanted tumor lung metastasis model was established,randomly divided into model group,Chinese medicine group,western medicine group,and combined medicine group.The changes in tumor growth and lung metastasis of mice in each group were observed.RT-PCR,Western Blot and immunohistochemical method was used to detect mRNA and protein expression of SDF-1/CXCR4 signaling pathway related molecules in lung tissue.Results:Four weeks after vaccination,the tumor formation rate and lung metastasis rate of mice were 100%.The inhibition rates of lung metastasis in the Chinese medicine group,the western medicine group and the combined medicine group were 42.85%,49.35%,and 52.59%,the three groups decreased than that in the model group(P<0.05).Compared with the model group,the mRNA expression of NF-κB,CXCR4,CXCL12,CD147 in the treatment groups significantly reduced(P<0.05,P<0.01),and the protein expression of CXCR4,SDF-1,NF-kB,p-NF-κB,CD147 in the treatment groups significantly reduced(except for CD147 protein in the Chinese medicine group)(P<0.01).Conclusion:Ru’ai Shuhou Formula can inhibit the occurrence of breast cancer lung metastasis,and reduce the expression of related molecules of SDF-1/CXCR4 signaling pathway in lung metastases.
