摘 要:目的:考察西洋参多糖(Panax quinquefolius polysaccharides,WQP)对正常大鼠肠道菌群的影响。方法:采用水提醇沉法制备WQP。将24只雄性Wistar大鼠随机分为正常组(Con组)和WQP低、中、高剂量组(WQPL、WQPM和WQPH组)。适应性饲喂7 d后,给药组大鼠灌胃不同剂量的WQP,正常组大鼠灌胃等体积生理盐水,分析大鼠生长状况、结肠组织结构和肠道菌群变化。结果:与Con组相比,WQP组对大鼠一般行为无不良影响,对大鼠体重无明显影响。与Con组相比,WQP组大鼠结肠的肠绒毛排列更为修长整齐、致密。门水平上,与Con组相比,不同剂量的WQP组厚壁菌门(Firmicutes)和变形菌门(Proteobacteria)的相对丰度减少,而拟杆菌门(Bacteroidetes)的相对丰度增加;科水平上,WQP可增加肠道菌群中乳酸杆菌科(Lactobacillaceae)和S24-7的相对丰度,降低瘤胃菌科(Ruminococcaceae)的相对丰度;属水平上,可增加乳杆菌属(Lactobacillus)的相对丰度,降低颤螺旋菌属(Oscillospira)和粪球菌属(Coprococcus)的相对丰度。结论:WQP可通过调节大鼠肠道菌群的组成和多样性,改善肠道菌群结构从而促进大鼠的肠道健康。Objective:To investigate the effect of Panax quinquefolius polysaccharides(WQP)on intestinal health of normal rats.Methods:Water-extraction alcohol-precipitation method was used to extract WQP.24 male Wistar rats were randomly divided into normal group(Con)and WQP with low dose(WQPL),middle dose(WQPM)and high dose groups(WQPH).After adaptive feeding for 7 days,rats in WQP group were given WQP with different doses,and normal rats were given normal saline with equal volume.The growth state,intestinal tissue structure and intestinal flora of rats were analyzed.Results:Compared with the Con group,WQP had no adverse effect on general behavior of rats,and it had no significant effect on body weight of rats.Compared with the Con group,the intestinal villi in the colon of the rats were more slender,neat and dense in WQP group.Compared with Con group,the relative abundance of Firmicutes and Proteobacteria decreased,while the relative abundance of Bacteroidetes increased in WQP group with different doses at phylum level.At the family level,the relative abundance of Lactobacillaceae and S24-7 in intestinal flora increased in WQP groups with different doses,but the relative abundance of Ruminococcaceae decreased.At the genus level,the relative abundance of Lactobacillus increased and the relative abundance of Oscillospira and Coprococcus decreased in WQP groups with different doses.Conclusion:WQP with different doses can improve the structure of intestinal flora by regulating the composition and diversity of intestinal flora in rats,so as to promote the intestinal health of normal rats.
