摘　　要：目的探讨结肠癌术后感染患者外周血中CK19 mRNA转录与蛋白表达的意义。方法选取2011年1月至2019年1月我院收治的结肠癌术后感染患者80例作为观察组,另选取同期我院收治的结肠息肉患者80例作为对照组。采用RT-PCR方法检测两组患者外周血中CK19 mRNA表达情况,使用ELISA检测两组患者外周血中CK19蛋白的表达情况。结果观察组CK19 mRNA阳性表达67例,阳性率为83.75%,对照组CK19 mRNA阳性表达22例,阳性率为27.50%,两组比较有显著性差异(P<0.01);处于C或D分期、低分化和有淋巴结转移患者的外周血CK19 mRNA转录与蛋白表达水平均分别显著高于A或B分期、中高分化和无淋巴结转移的患者(P<0.05或P<0.01)。结论结肠癌术后感染患者的外周血CK19 mRNA的表达阳性率较高,且其转录与蛋白表达水平与肿瘤的分期、分化程度及淋巴结转移具有明显相关性,可用于结肠癌病变的早期诊断。Objective To investigate the expression of CK19 mRNA and protein in peripheral blood of patients with postoperative colon cancer infection.Methods Eighty patients with postoperative colon cancer infection admitted to our hospital from January 2011 to January 2019 were enrolled as the observation group,and eighty patients with colon polyps admitted to our hospital were selected as the control group.The expression of CK19 mRNA in peripheral blood of the two groups was detected by RT-PCR,and the expression of CK19 protein in peripheral blood of the two groups was detected by ELISA.Results In the observation group,67 cases were positive for CK19 mRNA,the positive rate was 83.75%,and 22 cases were positive for CK19 mRNA in the control group,the positive rate was 27.50%,there was statistical significantly differences between the two groups(P<0.01).The levels of CK19 mRNA transcription and protein expression in peripheral blood of patients with C or D stage,poor differentiation and lymph node metastasis were significantly higher than those of patients with A or B stage,moderately high differentiation and no lymph node metastasis(P<0.05 or P<0.01).Conclusion The positive expression rate of CK19 mRNA in peripheral blood of patients with postoperative colon cancer infection is higher,and its transcription and protein expression levels are significantly correlated with tumor stage,differentiation and lymph node metastasis,and can be used for early diagnosis of colon cancer lesions.
