摘 要:目的研究沧源县蠓携带版纳病毒的全基因及其遗传进化特征。方法2018年在沧源县采集蠓,研磨后将上清液接种BHK-21细胞进行病毒分离的同时用宏转录组学方法检测蠓携带病毒情况,用Perl、Mafft等软件进行病毒序列比对、同源性和分子遗传进化分析。结果通过宏转录组学方法,从CYC1801组中获得1株版纳病毒的全基因序列,对全基因进行分析显示,节段4和节段7与越南株亲缘关系较近,其余节段均与中国株亲缘关系较近,根据节段12基因序列的系统进化分析,CYC1801与A2基因亚型处于同一进化分支,属于A2型版纳病毒。结论沧源县蠓携带有版纳病毒并可能作为传播媒介引起疾病流行,需加强监测及防控。Objective To study the whole genome and genetic evolution characteristics of the Banna virus carried by Culicoides in Cangyuan County.Methods Culicoides were collected in Cangyuan county in 2018.After grinding,the supernatant was inoculated onto BHK-21 cells for virus isolation.At the same time,the virus carrying status of Culicoides was detected by metatranscriptomics technology.Perl,Mafft and other bioinformatics softwares were used for sequence alignment,homology and molecular genetic evolution analysis.Results Through metatranscriptomics method,the whole genome of Banna virus CYC1801 strain was obtained.Analysis of the whole genome showed that segment 4 and segment 7 are closely related to the Vietnamese strains,other segments are closely related to China strains.According to the phylogenetic analysis of the segment 12 gene,CYC1801 and the A2 gene subtype are in the same evolutionary branch and belong to the A2 gene subtype of Banna virus.Conclusions Banna virus was carried by Culicoides collected in Cangyuan county,which may cause disease epidemic as a vector,so monitoring and prevention should be strengthened.
