摘　　要：目的探讨Fosb蛋白表达与癫癎的关系。方法雄性SD大鼠70只,随机分为对照组(n=9)和模型组(n=61)。采用戊四唑(PTZ)诱导大鼠癫癎模型,模型组再依据PTZ注射次数和癫癎发作分级分为8次4级亚组、16次2级亚组和16次4级亚组;取大鼠海马组织进行mRNA测序,并对测序结果建立Network作用网络。另取海马组织行苏木精-伊红染色,提取目标基因总RNA行qPCR检测;免疫组化检验Fosb蛋白表达水平。结果Fosb、Gadd45g、Npas4、Atf 34个基因变化与PTZ注射次数及癫癎发病程度呈正相关,具有相互关联性。Atf 3:8次4级亚组与对照组比较(P=3.97E-17);16次2级亚组与对照组比较(P=6.85E-06);16次4级亚组与对照组比较(P=4.88E-84)。Fosb:8次4级亚组与对照组比较(P=0.0000);16次2级亚组与对照组比较(P=1.81E-176);16次4级亚组与对照组比较(P=0.0000)。Gadd 45g:8次4级亚组与对照组比较(P=6.81E-151);16次2级亚组与对照组比较(P=6.49E-39);16次4级与对照组比较(P=0.0000)。Npas:8次4级亚组与对照组比较(P=0.0306);16次2级亚组与对照组比较(P=0.0430);16次4级亚组与对照组比较(P=0.0080)。qPCR检测显示:Fosb和Nasp4蛋白水平明显高于对照组(Fosb:8次4级亚组与对照组比较,P=0.0462;16次4级亚组与对照组比较(P=0.0193);Npas4:8次4级亚组与对照组比较(P=0.0306);16次2级亚组与对照组比较(P=0.0430);16次4级亚组与对照组比较(P=0.0080)。免疫组化结果显示,模型组Fosb蛋白含量较对照组明显增加(P=0.0004)。结论Fosb蛋白表达水平与癫癎发病密切相关。Aim To investigate the relationship of the expression of Fosb protein and epilepsy.Methods Pentylenetet was used to induce the epileptic model in rats.mRNA sequencing was performed in hippocampal tissues,and a network was established based on the sequencing results.Brain tissue was taken for HE staining,and total RNA was extracted for Q-PCR detection and immunohistochemical experiment of the target gene.Results The changes of Fosb,Gadd 45g,Npas 4 and Atf 3 were positively correlated and correlated with the times of PTZ injection and the seizure degree of epilepsy.Atf 3:PTZ 8 times 4 grade subgroup compared with the control group(P=3.97E-17);PTZ 16 times 2 grade subgroup compared with the control group(P=6.85E-06);PTZ16 times 4 grade subgroup compared with the control group(P=4.88E-84).Fosb:PTZ 8 times 4 grade subgroup compared with the control group(P=0.0000);PTZ 16 times 2 grade subgroup vs.the control group(P=1.81E-176);PTZ16 times 4 grade subgroup compared with the control group(P=0.0000).Gadd 45g:PTZ 8 times 4 grade subgroup compared with the control group(P=6.81E-151);PTZ 16 times 2 grade subgroup vs.the control group(P=6.49E-39);PTZ 16 times 4 grade subgroup vs.the control group(P=0.0000).Npas:PTZ 8 times 4 grade subgroup compared with the control group(P=0.0306);PTZ 16 times 2 grade subgroup compared with the control group(P=0.0430);PTZ16 times 4 grade subgroup compared with the control group(P=0.0080).Q-PCR results showed that Fosb and Nasp4 were significantly higher than the normal group Fosb:PTZ 8 times 4 grade subgroup compared with the control group(P=0.0462);PTZ16 times 4 grade subgroup compared with the control group(P=0.0193).Npas:PTZ 8 times 4 grade subgroup compared with the control group(P=0.0306);PTZ 16 times 2 grade subgroup compared with the control group(P=0.0430);PTZ16 times 4 grade subgroup compared with the control group(P=0.0080).Immunohistochemical results showed that the content of Fosb in the model group was significantly increased compared with the normal group(P=0.0004).Conclusion F
