摘 要:以东亚飞蝗为原料提取多肽,利用动物试验验证其多肽对高脂血症的抑制作用,为昆虫资源开发提供新的思路和理论依据。采用酶解提取法从东亚飞蝗中分别提取多肽,利用正交试验,采用单因素考察等方式,设置四因素三水平对东亚飞蝗多肽提取工艺进行优化。随后利用东亚飞蝗多肽对大鼠进行降血脂试验,采用5周龄SPF级SD雄性大鼠40只,分别设置正常饲料组(正常对照组CON)、模型组(HFD组)、阳性对照组(氟伐他汀钠组AC)、多肽试验干预组(PE8),喂食4周。结果表明:多肽提取最佳工艺为反应时间3 h,反应温度40℃,料液比1∶8,酶质量分数0.3%。动物试验经过东亚飞蝗多肽干预后,血清中总胆固醇(P<0.01)、甘油三酯(P<0.01)、低密度脂蛋白(P<0.01)、高密度脂蛋白(P<0.05)显著降低。综上,东亚飞蝗多肽可以改善高脂血症大鼠症状,并通过改善脂肪代谢紊乱,减轻肝脏脂肪堆积,达到对高脂血症的预防效果。The polypeptide was extracted from Locusta migratoria manilensis and its inhibitory effect on hyperlipidemia was verified by animal experiments,which would provide new ideas and theoretical basis for the development of insect resources.The polypeptide was extracted from Locusta migratoria manilensis by enzymatic hydrolysis,and the orthogonal test and the single-factor inspection were used to set four factors and three levels to optimize the extraction process of polypeptide from Locusta migratoria manilensis.Subsequently,the hypolipidemic test was carried out on the rats with the locust polypeptide,and 40 male SPF SD rats of five weeks old were fed with normal feed group(normal control group CON),model control group(HFD group),positive control group(fluvastatin sodium group AC)and polypeptide test intervention group(PE8)for four weeks.The results show that the optimal process for polypeptide extraction is reaction time 3 hours,reaction temperature 40℃,feed-liquid ratio 1∶8 and enzyme concentration 0.3%.In animal experiments,after intervening with the polypeptide,polypeptide from Locusta migratoria manilensis,serum TC(P<0.01),TG(P<0.01),LDL-C(P<0.01)and HDL-C(P<0.05)were significantly reduced.In summary,polypeptide from Locusta migratoria manilensis can improve hyperlipidemia rats and reduce liver fat accumulation by improving fat metabolism disorders so as to achieve the preventive effect on hyperlipidemia.Therefore,polypeptide from Locusta migratoria manilensis can be further developed as a health product to prevent hyperlipidemia.
