摘 要:为防治奶牛产后子宫内膜炎提供益生菌株,从健康奶牛生殖道分离筛选益生乳酸菌。利用子宫采样器采集28头产后45-65 d健康奶牛子宫内容物,通过体外培养、分离和纯化,结合形态、生理生化特征和16S rDNA序列进行鉴定,并检测菌株的抗菌活性和物质、抗生素敏感性及细胞黏附性。结果表明:共分离筛选出15株乳酸菌,分别为唾液乳杆菌7株,粪肠球菌4株,殊异肠球菌、海氏肠球菌、耐久肠球菌和绒毛肠球菌各1株。15株乳酸菌均具有较好的抗菌活性和抗生素敏感性;殊异肠球菌、海氏肠球菌、耐久肠球菌和唾液乳杆菌主要抗菌物质为有机酸;绒毛肠球菌和粪肠球菌主要抗菌物质除有机酸外还分泌细菌素。殊异肠球菌和绒毛肠球菌对奶牛子宫内膜上皮细胞具有较强黏附能力。殊异肠球菌和绒毛肠球菌可作为潜在的益生菌,用于开发防治奶牛子宫内膜炎的微生态制剂。The aim of the study was to isolate lactic acid bacteria(LAB)strains to provide probiotic strains for preventing and treating endometritis of postpartum cows.The uterine secretions of28 healthy dairy cows from 45 to 65 d after parturition were collected using a uterine sampler.They were cultured,isolated and purified in vitro,and identified in combination with morphology,physiological and biochemical characteristics and 16S rDNA sequence.The LAB isolates were tested for anti-bacteria activity and substances,drug resistance,and adhesion to endometrial epithelial cells.The results show that a total of 15 strains of LAB were isolated and identified,including 7 strains of Lactobacillus salivary,4 strains of Enterococcus faecium,and 1 strain each of Enterococcus disbar,Enterococcus hire,Enterococcus durans and Enterococcus villorum.Fifteen strains of LAB have good antibacterial activity and antibiotic sensitivity.The main antimicrobial substances of Enterococcus disbar,Enterococcus hire,Enterococcus durans and Lactobacillus salivary were organic acids.However,the main antimicrobial substances of Enterococcus villorum and Enterococcus faecium were organic acids and bacteriocin.Enterococcus disbar and Enterococcus villorum had strong adhesion ability to endometrial epithelial cells of cow.The results indicate that Enterococcus disbar and Enterococcus villorum have the potential as probiotics to further develop Lactobacillus microecological preparations.
