慢病毒介导eIF3b-shRNA对裸鼠MCF-7乳腺癌移植瘤的抑制作用

摘　　要：目的:探讨慢病毒介导eIF3b-shRNA对裸鼠乳腺癌移植瘤生长、增殖及转移的影响。方法:利用慢病毒介导eIF3b-shRNA转染MCF-7乳腺癌细胞,建立裸鼠乳腺癌模型,裸鼠分成三组,每组5只:I组(接种转染eIF3b-shRNA的MCF-7乳腺癌细胞)、Ⅱ组(接种转染空载体的MCF-7乳腺癌细胞)和Ⅲ组(接种正常MCF-7乳腺癌细胞)。观察制瘤成功后裸鼠肿瘤体积生长、HE染色镜下观察移植瘤组织和肺组织,并应用实时荧光定量PCR法检测eIF3b mRNA表达,Western blot印迹法检测eIF3b、增殖标记蛋白Ki-67及迁移标记蛋白MMP-9的表达。结果:三组裸鼠成瘤后,不同时间点对三组裸鼠进行观察,Ⅰ组肿瘤体积均明显小于Ⅱ组与Ⅲ组(P<0.05),而Ⅱ组与Ⅲ组之间比较,差异无统计学意义(P>0.05)。实时荧光定量PCR法检测结果显示:I组裸鼠移植瘤组织中eIF3b mRNA相对表达量明显下降,明显低于Ⅱ组和Ⅲ组,差异具有统计学意义(P<0.05),而Ⅱ组和Ⅲ组之间eIF3b mRNA相对表达量比较,差异无统计学意义(P>0.05)。Western blot印迹法检测结果显示:Ⅰ组裸鼠移植瘤组织中eIF3b、增殖标记蛋白Ki-67和迁移标记蛋白MMP-9相对表达量较Ⅱ组和Ⅲ组显著下降,差异具有统计学意义(P<0.05),而Ⅱ组和Ⅲ组之间各蛋白相对表达量比较,差异无统计学意义(P>0.05)。解剖三组裸鼠,大体肉眼观察发现Ⅱ组和Ⅲ组的肺组织切开可见多灶灰白小结节,而Ⅰ组裸鼠肺组织中未找到明显结节。镜下观察发现,Ⅱ组和Ⅲ组裸鼠肺泡组织中灰白小结节为腺管样异型细胞呈结节样生长,而Ⅰ组镜下见肺泡组织结构正常,未找到转移灶。结论:慢病毒介导下调eIF3b表达对乳腺癌细胞在裸鼠体内的移植瘤生长、增殖及肺转移有明显抑制作用。Objective:To investigate the effect of lentivirus mediated eIF3b-shRNA on growth,proliferation and metastasis of breast cancer xenografts in nude mice.Methods:MCF-7 breast cancer cells were transfected by lentivirus mediated eIF3b-shRNA,and a nude mouse breast cancer model was established.Nude mice were divided into three groups,5 nude mice in each group:group Ⅰ(MCF-7 transfected breast cancer cells transfected with eIF3b-shRNA),Ⅱgroup(inoculated with MCF-7 breast cancer cells transfected with empty carrier)andⅢgroup(inoculated normal MCF-7 cells).The tumor volume growth of nude mice after successful tumor preparation was observed.The transplanted tumor tissue and lung tissue were observed under HE staining microscope.The gene expression of eIF3b was detected by real-time fluorescence quantitative PCR,and the expression of eIF3b,proliferation marker protein Ki-67 and migration marker protein MMP-9 were detected by Western blot.Results:The tumor volume of group I was significantly smaller than that of groupⅡand groupⅢ(P<0.05),but there was no significant difference between groupⅡand groupⅢ(P>0.05).The results of real-time fluorescence quantitative PCR showed that the relative expression of eIF3b mRNA in transplanted tumor tissues of nude mice in group I decreased significantly,which was significantly lower than that in groupⅡandⅢ(P<0.05),while there was no statistical significance in the comparison of the relative expression of eIF3b mRNA between groupⅡandⅢ(P>0.05).The results of Western blot showed that the relative expression of eIF3b,proliferation marker protein Ki-67 and migration marker protein MMP-9 in group I was significantly lower than that in groupⅡand groupⅢ(P<0.05),but there was no significant difference between groupⅡand groupⅢ(P>0.05).After dissecting the three groups of nude mice,it was found that multiple gray and small nodules could be seen in the lung tissue of groupⅡand groupⅢ,while no obvious nodules were found in the lung tissue of group I.Microscopic observat
