摘 要:目的:探讨长链非编码RNA(long non-coding RNA,lncRNA)ALMS1-IT1在结肠癌组织中的表达及其临床意义,并探索其对结肠癌细胞增殖和侵袭的影响。方法:应用GEPIA平台分析TCGA数据库中ALMS1-IT1在结肠癌与正常组织中的表达水平及其与患者预后的相关性。通过实时荧光定量PCR(real-time quantitative PCR,RT-qPCR)检测ALMS1-IT1在结肠癌及癌旁组织中的表达并分析其与临床病理特征的相关性。pcDNA3.1-ALMS1-IT1和ALMS1-IT1 siRNA分别转染HCT116细胞,RT-qPCR检测转染效率,CCK-8、EdU和Transwell小室检测HCT116细胞增殖和侵袭能力。结果:TCGA数据提示,ALMS1-IT1在结肠癌组织中表达显著高于正常组织,我们的数据也证实ALMS1-IT1在结肠癌组织中显著高表达。TCGA数据生存分析发现ALMS1-IT1高表达的结肠癌患者总生存率和无病生存率均显著降低。干扰ALMS1-IT1显著抑制HCT116细胞增殖和侵袭,而过表达ALMS1-IT1则明显增强结肠癌细胞增殖和侵袭。结论:ALMS1-IT1在结肠癌组织中表达上调,其高表达提示患者预后不良,ALMS1-IT1通过促进结肠癌细胞增殖和侵袭发挥癌基因功能。Objective:To clarify the expression of ALMS1-IT1 in colon cancer tissues and its clinical significance,and to explore its effects on colon cancer cell proliferation and invasion.Methods:The GEPIA platform was used to analyze the expression levels of ALMS1-IT1 in colon cancer and normal tissues and its correlation with the prognosis of patients in the TCGA database.The expression of ALMS1-IT1 in colon cancer and tumor-adjacent tissues was detected by real-time quantitative PCR(RT-qPCR)and its correlation with clinicopathological characteristics was analyzed.The pcDNA3.1-ALMS1-IT1 and ALMS1-IT1 siRNA were transfected into HCT116 cells,respectively.The transfection efficiency was detected by RT-qPCR.The proliferation and invasion of HCT116 cells were measured by CCK-8,EdU and Transwell assays.Results:TCGA data suggested that the expression of ALMS1-IT1 in colon cancer tissues was significantly higher than that in normal tissues.Our data also confirmed that ALMS1-IT1 was significantly overexpressed in colon cancer tissues.Survival analysis of TCGA data found that the overall survival and disease-free survival rates of colon cancer patients with high ALMS1-IT1 expression were significantly reduced.Knockdown of ALMS1-IT1 significantly inhibited the proliferation and invasion of HCT116 cells,while overexpression of ALMS1-IT1 markedly enhanced colon cancer cell proliferation and invasion.Conclusion:ALMS1-IT1 was up-regulated in colon cancer tissues,and its high expression indicated poor prognosis of patients.ALMS1-IT1 exerted an oncogenic function by promoting colon cancer cell proliferation and invasion.
