摘　　要：[目的]探讨洋甘菊提取物对高血压大鼠的降压及对肾脏的保护作用。[方法]建立两肾一夹高血压大鼠模型,随机分为模型组、洋甘菊提取物(157.5 mg/kg)和(315 mg/kg)剂量组及卡托普利(25 mg/kg)阳性对照组。另取8只大鼠作为假手术组,假手术组和模型组每天给予等量的生理盐水,连续灌胃给药4 w,每周测量血压。检测大鼠血清尿素氮和肌酐水平,ELISA法检测大鼠血清中白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的含量;采用HE染色观察各组肾脏组织的形态学变化。[结果]给药后,洋甘菊提取物(157.5 mg/kg)、(315 mg/kg)剂量组和卡托普利组收缩压分别为131.20±5.77 mmHg、128.92±13.62 mmHg和127±8.86 mmHg,明显低于模型组(P<0.05)。洋甘菊提取物(157.5 mg/kg)剂量组血清IL-6为4.24±1.31 ng/mL,明显低于模型组(P<0.05);BUN、CR和IL-10含量无明显变化(P>0.05);洋甘菊提取物各剂量组中部分肾小球萎缩、肾小管炎性浸润状况得到缓解。[结论]洋甘菊提取物能降低高血压大鼠血压(131.20±5.77 mmHg、128.92±13.62 mmHg)、IL-6(4.24±1.31 ng/mL)含量并减轻肾脏损伤的作用,其可能通过降低IL-6含量从而改善炎症反应,起到降压和保护肾脏的作用。[Objective]To study the effects of Matricaria chamomilla extract on blood pressure and renal tissue renal hypertensive rats.[Method]Two-kidney one-clip(2K1C)model of renovascular hypertension,The rats were randomly divided into groups:model group,(157.5 mg/kg)and(315 mg/kg)dose groups of Matricaria chamomilla extract;captopril positive drug group(25 mg/kg);and another six normal rats were selected as sham operation group.the sham operation group and the model group were given the same amount of normal saline,each group was given a continuous gavage for four weeks,and the arterial blood pressure was measured once a week,After four weeks of treatment,blood was collected from the rat abdominal aorta for determination of serum IL-6 and IL-10 level,detection of serum creatinine and urea nitrogen in rats by automatic biochemical analyzer.and renal pathological changes were observed by HE staining.[Result]After treatment,Matricaria chamomilla extract and captopril positive drug group showed obvious hypotensive effect,the systolic blood pressure were 131.20±5.77 mm-Hg,128.92±13.62 mmHg and 127±8.86 mmHg,(P<0.05).The contents of IL-6 in Matricaria chamomilla extract(157.5 mg/kg)dose group was 4.24±1.31 ng/mL lower than the model group(P<0.05),The contents of BUN,CR and IL-10 in Matricaria chamomilla extract groups had no significant difference(P>0.05);atrophy of glomeruli and inflammatory cell infiltration of renal tubular mesangial alleviated.[Conclusion]Matricaria chamomilla extract is effective in decreasing hypertension blood pressure,effectively protect renal function and improve the level of renal fibrosis.It may be related to the decrease of IL-6 content,improve inflammatory response to decrease hypertension blood pressure and protect renal.
