摘 要:目的探讨细胞维甲酸结合蛋白2(CRABP2)在食管鳞癌中的表达,观察联合全反式维甲酸(ATRA)对食管鳞癌细胞的作用。方法基于GEO数据库分析CRABP2在食管鳞癌中的表达情况。收集新疆医科大学第一附属医院病理科2008年1月—2019年6月诊断的240例食管鳞癌患者组织芯片,采用免疫组织化学法检测CRABP2在食管鳞癌患者组织样本中的表达情况。通过Western blot、CCK-8、Transwell和荧光免疫等观察CRABP2及联合ATRA对食管鳞癌细胞增殖、侵袭和迁移的影响。结果GSE20347数据集分析结果显示,CRABP2在食管鳞癌组织中的表达显著低于食管正常组织,是食管鳞癌的差异表达基因,差异倍数log2FC=-3.19。CRABP2在正常食管黏膜组织中的阳性表达率为100.0%,在食管鳞癌组织中的阳性表达率为75.6%,差异有统计学意义(x^(2)=47.84,P=0.000)。CRABP2、ATRA均可抑制KYSE150细胞的增殖、迁移和侵袭;CRABP2与ATRA联合使用较二者单独使用更能增加CRABP2的表达,抑制KYSE150细胞发生增殖、迁移和侵袭的效果更为显著。结论CRABP2具有抑制食管鳞癌增殖、迁移的作用,联合应用ATRA能进一步增强抑制食管鳞癌细胞增殖、侵袭、迁移的作用。Objective To explore the expression of cellular retinoic acid binding protein 2(CRABP2)in esophageal squamous cell carcinoma,and to observe the effect of combined trans retinoic acid(ATRA)on esophageal squamous cell carcinoma.Methods The expression of CRABP2 in esophageal squamous cell carcinoma was analyzed by GEO database.The tissue chips of 240 patients with esophageal squamous cell carcinoma diagnosed in the hospital from January 2008 to June 2019 were collected,and the expression of CRABP2 in esophageal squamous cell carcinoma tissues were detected by immunohistochemistry.The effects of CRABP2 and ATRA on proliferation,invasion and migration of esophageal squamous cell carcinoma were observed by cell function experiments.Results GSE20347 data set analysis showed that the expression of CRABP2 in esophageal squamous cell carcinoma was significantly lower than that in normal esophageal tissues,and it was a different expressed gene in esophageal squamous cell carcinoma with the difference multiple of log2FC=-3.19.The positive expression rate of CRABP2 in normal oesophageal mucosal tissue was 100.0%and 75.6%in esophageal squamous cell carcinoma,which was statistically significant(x^(2)=47.84,P=0.000).CRABP2 and ATRA could inhibit the proliferation,migration and invasion of KYSE150 cell.Compared with the single use of CRABP2 and ATRA,the combination of CRABP2 and ATRA were increased with the expression of CRABP2 and significantly inhibited proliferation,migration and invasion of KYSE150.Conclusion CRABP2 can not only inhibit proliferation and migration of esophageal squamous cell carcinoma by itself,but also can further promote the inhibition effect of CRABP2 combined with ATRA.
