摘  要:目的:通过研究亚慢性铝暴露对大鼠记忆功能及组蛋白乙酰基转移酶1(HAT1)表达的影响,探究铝损伤记忆功能的机制。方法:将Wistar大鼠随机分为对照组(control)和低染毒组(Al-L)、中染毒组(Al-M)、高染毒组(Al-H),以出生当天(1 d)至90 d持续染毒方式建立亚慢性铝暴露大鼠模型。用Morris水迷宫检测大鼠的记忆功能,用尼氏染色法观察海马CA1区神经细胞的形态改变,real time RT-PCR检测大鼠海马HAT1 mRNA的表达,Western Blot检测大鼠海马HAT1蛋白的表达。结果:与对照组相比,染铝组大鼠在目标象限停留时间较短,穿越平台次数较少,表明大鼠空间记忆能力受损;尼氏染色显示大鼠海马CA1区神经细胞形态受损,同时大鼠海马HAT1 mRNA和蛋白表达均降低。结论:亚慢性铝暴露降低大鼠海马CA1区HAT1表达,可能导致大鼠空间记忆功能障碍。Objective: To explore the mechanism of aluminum damage to memory function by studying the effects of subchronic aluminum exposure on memory function and the expression of histone acetyltransferase 1(HAT1) in rats. Methods: Wistar rats were divided into a control group and three experiment groups in which rats were exposed to aluminum with different doses, low dose group(Al-L), middle dose group(Al-M), and high dose group(AL-H) respectively. The subchronic aluminum exposure rat model was established by continuous exposure from the partal day 1 d to 90 d. The Morris water maze was used to test the memory function of the rats, and Nissl staining was used to observe the morphological changes of neuronal cells in hippocampal CA1 region. The mRNA expression of HAT1 in hippocampus of the rats was detected by real time RT-PCR, and the protein expression of HAT1 in hippocampus of the rats was detected by Western Blot. Results: Compared with the control group, the rats in the aluminum exposure group had shorter time spend in the target quadrant and fewer crossing times, which indicated that the spatial memory ability of rats was damaged. Nissl staining showed that the morphology of neuronal cells in hippocampal CA1 region of rats was damaged, at the same time, the expression levels of hippocampal HAT1 mRNA and protein were decreased in rats. Conclusion: Subchronic aluminum exposure reduces the expression of HAT1 in hippocampal CA1 region, which may lead to spatial memory dysfunction in rats.
