TLR5基因单核苷酸多态性与幽门螺杆菌感染、非贲门胃癌发病风险的关联

摘　　要：目的用病例对照关联研究方法,在包头汉族人群中检测Toll样受体(TLR)5基因单核苷酸多态性与幽门螺杆菌感染、非贲门胃癌发病风险的关系。方法采用TaqMan法对288例非贲门胃癌患者(病例组)及年龄、性别与其匹配的281例体检者(正常对照组)进行基因分型;用酶联免疫吸附试验检测正常对照组中幽门螺杆菌的感染情况;使用Haploview软件构建单体型,同时运用非条件性Logistic回归计算比值比(OR)及其95%可信区间(CI),评估各等位基因、基因型及单体型与幽门螺杆菌感染、非贲门胃癌发病风险的关系。结果在正常对照组中,幽门螺旋杆菌的感染率为43.3%,TLR5基因SNP rs5744138、rs2241096、rs160827、rs2096141、rs160812位点的等位基因、基因型与幽门螺杆菌感染、非贲门胃癌的发病风险无关联(P>0.05)。同时各单体型与幽门螺杆菌感染、非贲门胃癌的发病风险无统计学意义(P>0.05)。结论TLR5基因多态性可能在幽门螺旋杆菌感染、非贲门胃癌的发病风险中不起主要作用。Objective To investigate the relationship between single nucleotide polymorphisms(SNPs)in TLR5 gene with Helicobacter(H.)pylori infection and non-cardia gastric cancer risk by case-control association study in Baotou Han population.Methods SNPs were genotyped by TaqMan method among 288 cases with non-cardia gastric cancer and 281 normal controls.The enzyme-linked immunosorbent assay(ELISA)was used to detect H.pylori infection.Haplotypes were constructed by Haploview software.Odds ratios(ORs)and 95%confidence intervals(CIs)were calculated by Logistic regression analysis to evaluate the correlations of the alleles,genotypes,and haplotypes with H.pylori infection and non-cardia gastric cancer risk.Results In normal controls,the rate of H.pylori infection was 43.3%.The allele and genotype of TLR5 SNP rs5744138,rs2241096,rs160827,rs2096141,rs160812 were not associated with the risk of H.pylori infection and non-cardia gastric cancer(P>0.05).There was no significant difference between haplotypes and the risk of H.pylori infection and non-cardia gastric cancer(P>0.05).Conclusions The SNP in TLR5 gene might not play a role in the H.pylori infection and non-cardia gastric cancer risk.
