摘  要:目的探讨非洲爪蟾驱动蛋白样蛋白2靶蛋白(TPX2)在乳腺癌与癌旁组织中的差异性表达及其与其他临床病理指标的关系, 分析TPX2表达与患者生存率的相关性。方法采用免疫组织化学检测广州医科大学附属第一医院115例乳腺癌组织及69例癌旁组织中TPX2的表达, 比较两组TPX2表达率的差异, 以及TPX2的表达同其他临床病理指标的关系。采用Kaplan-Meier生存曲线法进行生存分析, 采用COX比例风险回归模型分析影响预后的危险因素。结果乳腺癌组织中TPX2的阳性表达率高于癌旁组织(54.8%比11.6%, χ^(2)=9.358, P<0.05)。TPX2的表达与肿瘤较低的分化程度(P<0.05)及ER表达(P<0.05)明显相关, 与患者年龄、TNM分期、肿瘤大小、淋巴结转移、孕激素受体(PR)、人表皮生长因子受体2(Her-2)表达无明显相关。Kaplan-Meier法生存分析显示, TPX2蛋白表达水平高的患者预后较低表达的患者差, 差异有统计学意义(P<0.05)。Cox比例风险回归模型分析显示, 雌激素受体(ER)与TPX2是乳腺癌患者的独立预后因子。结论 TPX2在乳腺癌组织中的表达水平明显升高, 且TPX2高表达的乳腺癌患者表现出较差预后, 表明TPX2可能促进乳腺癌的发生与发展, 并可作为潜在的治疗靶点。Objective To detect the expression of targeting protein for Xenopus kinesin-like protein 2(TPX2)in breast cancer tissue and adjacent noncancerous tissues,investigate the correlation between TPX2 and other clinical features,and to evaluate the prognostic significance of TPX2.Methods The expression of TPX2 was detected by immunohistochemical method in 115 breast cancer patients and 69 adjacent noncancerous tissues from the First Affiliated Hospital of Guangzhou Medical University.We compared TPX2 expression level between the two groups and investigated the relationship between TPX2 expression and other clinical features.Results The expression of TPX2 in breast cancer tissues was significantly higher than in adjacent noncancerous tissues(45.83%vs.20.75%,χ^(2)=9.358,P<0.05).The TPX2 expression level showed close relationship with grade(P<0.05)and estrogen receptor(ER)expression(P<0.05),but no statistical correlation with age,TNM stage,tumor size,lymph node metastasis,progesterone receptor(PR)and human epidermal growth factor receptor-2(Her-2)expression.The Kaplan-Meier survival analysis showed that patients with high TPX2 expression level had poor prognosis(P<0.05).The Cox regression analysis showed that the expression of ER and TPX2 was the independent prognostic factors for breast cancer.Conclusion The expression of TPX2 is obviously increased in breast cancer tissues.Patients with high TPX2 expression level had significantly poor prognosis.The results may reveal that TPX2 may promote the development of breast cancer and may be a potential therapeutic target.
