摘　　要：目的检测NF-κB在粪肠球菌感染的慢性根尖周炎中的作用。方法比对10例临床再治疗慢性根尖周炎组织和10例健康根尖周组织,通过real-time PCR对NF-κB的表达情况进行比较。建立粪肠球菌感染的大鼠根尖周炎动物模型。右下颌第一磨牙开髓,随机分为两组,A组20只,为未封菌组;B组20只,为封菌组,两组动物按照处理时间(周)分为4个亚组。对照组为对侧第一磨牙。采用免疫组化和real-time PCR检测NF-κB的表达情况。结果慢性根尖周炎组织NF-κB水平显著高于健康组织(t=6.123,P<0.001)。HE染色确认A组及B组大鼠根尖周炎模型建立成功。real-time PCR结果显示A组及B组NF-κB的基因表达水平趋势一致,2周时表达量最高(F=37.824,P<0.001;F=39.094,P<0.001)。免疫组化结果显示两组NF-κB蛋白表达水平在2周时最高(F=357.031,P<0.001;F=398.243,P<0.001)。B组NF-κB基因在2周及3周时表达量高于A组(t=10.464,P=0.002;t=12.093,P=0.013)。B组NF-κB蛋白在2周及3周时表达量高于A组(t=11.093,P<0.001;t=10.324,P=0.023)。结论NF-κB参与了粪肠球菌感染根尖周炎的发生与发展过程。Objective To explore the role of NF-κB in chronic periapical periodontitis caused by Enterococcus faecalis.Methods The expression of NF-κB in 10 clinical patients with chronic apical periodontitis and in 10 individuals with healthy periapical tissues were compared using real-time PCR.Animal models of rat periapical periodontitis caused by Enterococcus faecalis were established.The right mandibular first molars were pulped and randomly divided into two groups,20 in group A(unsealed group)and 20 in group B(sealed group).The two groups of animals were divided into 4 subgroups according to the number of weeks.The contralateral first molar was the control group.Immunohistochemistry and real-time PCR were used to detect the expression of NF-κB.Results There was significant difference in NF-κB between chronic periapical periodontitis and healthy tissues(t=6.123,P<0.001).The HE confirmed that the animal models of periapical periodontitis in groups A and B were successfully established.real-time PCR showed that the gene expression levels of NF-κB in group A and group B had the same trend,and the expression was the highest at 2 weeks(F=37.824,P<0.001;F=39.094,P<0.001).The immunohistochemistry also showed that the protein expression level of NF-κB was the highest at 2 weeks(F=357.031,P<0.001;F=398.243,P<0.001).The gene and protein expression of NF-κB in group B was higher than that in group A at 2 and 3 weeks(all P<0.05).Conclusion NF-κB was closely related to the occurrence and development of periapical periodontitis caused by Enterococcus faecalis.
