摘 要:目的:探讨藏红花提取液对兔慢性高眼压视网膜神经节细胞损伤的保护作用。方法:将新西兰白兔随机分为对照组、高眼压组、治疗Ⅰ组和Ⅱ组,每组5只。高眼压组和治疗Ⅰ、Ⅱ组的兔眼前房注入3 g/L复方卡波姆溶液制作慢性高眼压模型,对照组仅作前房穿刺。治疗Ⅰ、Ⅱ组动物每日耳缘分别静注藏红花提取液20、80 mg/kg;高眼压组和对照组各注射2 ml 0.9%氯化钠溶液,连续28 d。然后所有动物经心脏灌流固定,取球后视神经和视网膜,视神经包埋后作超薄切片,电镜下观察节细胞轴突;视网膜视神经作冰冻切片,进行HE染色、TUNEL染色及免疫组化检测。结果:高眼压组视网膜各层细胞排列紊乱,节细胞数目减少,神经纤维层明显变薄,轴突数目显著减少;治疗组明显改善,而治疗Ⅱ组视网膜形态结构接近对照组,各层结构层次清楚,细胞排列整齐,细胞数量基本正常。高眼压组视网膜中的凋亡节细胞及Caspase 3阳性细胞数量最多,注射藏红花提取液后凋亡的节细胞及Caspase 3阳性细胞数目明显减少,在治疗Ⅱ组仅见个别凋亡的节细胞及偶见Caspase 3阳性细胞。结论:慢性高眼压可导致兔眼视网膜神经节细胞及轴突结构损害、视网膜节细胞凋亡;藏红花提取液可通过减少节细胞凋亡、降低Caspase 3表达,从而保护视网膜神经节细胞。Objective:To investigate the protective effect of crocus sativus extract on the retinal ganglion cells injury in the rabbits with chronic ocular hypertension(OHT).Methods:Twenty rabbits were randomly divided into control group,OHT group,treatment groupⅠandⅡ,with 5 rabbits in each group.OHT model was induced by injection of 3 g/L carbomer into anterior chamber in the OHT group and two treatment groups,while anterior chamber paracentesis without injection was performed in the control group.Rabbits in treatment groupⅠandⅡwere intravenously injected with crocus sativus extract 20 and 80 mg/kg respectively,while rabbits in the OHT and control groups were injected with 2 ml of 0.9%sodium chloride solution for 28 consecutive days.Then all the rabbits were sacrificed by perfusion and optic nerves behind the eyeball and retina slices were collected.The frozen slices were examined by a conventional transmission electron microscopy and the apoptosis of retinal ganglion cell was detected by immuno-histochemistry,HE and TUNEL staining.Results:In the OHT group,the arrangement of cells in each layer of retina was disordered,the number of ganglion cells decreased,the nerve fiber layer became thinner,and the number of axons decreased significantly.The treatment groups were significantly improved,while the retinal morphology of treatment groupⅡwas close to the control group,with clear levels of each layer structure,neat arrangement of cells and normal number of cells.In the OHT group,the number of apoptotic ganglion cells and Caspase 3 positive cells in retina was the highest,and decreased significantly after injection of crocus sativus extract,and only individual apoptotic ganglion cells and occasionally Caspase 3 positive cells were found.Conclusion:Chronic OHT can lead to damage of retinal ganglion cells and axon structure and apoptosis of retinal ganglion cells in rabbit eyes.Crocus sativus extract can protect retinal ganglion cells by reducing ganglion cell apoptosis and Caspase 3 expression.
